Calcium Signaling: Septins Organize the SOC Channel

چکیده

Store-operated Ca(2+) entry is a widely encountered mechanism for generating cytoplasmic Ca(2+) signals. From a whole-genome screen in HeLa cells, a new study reveals that cytoskeletal septins play a critical role in this process, by organizing signaling structures in the plasma membrane.

Store-operated Ca2+ entry is exaggerated in fresh preglomerular vascular smooth muscle cells of SHR.

BACKGROUND Regulation of preglomerular vasomotor tone vessels ultimately control glomerular filtration rate, sodium reabsorption and systemic blood pressure. To gain insight into the complex renal hemodynamic factors that may result in hypertension, we studied calcium signaling pathways. METHODS Fresh, single, preglomerular vascular smooth muscle cells (VSMC) were isolated from 5- to 6-week-o...

Structurally Distinct Ca2+ Signaling Domains of Sperm Flagella Orchestrate Tyrosine Phosphorylation and Motility

Spermatozoa must leave one organism, navigate long distances, and deliver their paternal DNA into a mature egg. For successful navigation and delivery, a sperm-specific calcium channel is activated in the mammalian flagellum. The genes encoding this channel (CatSpers) appear first in ancient uniflagellates, suggesting that sperm use adaptive strategies developed long ago for single-cell navigat...

TRPC1: a core component of store-operated calcium channels.

The TRPC (transient receptor potential canonical) proteins are activated in response to agonist-stimulated PIP(2) (phosphatidylinositol 4,5-bisphosphate) hydrolysis and have been suggested as candidate components of the elusive SOC (store-operated calcium channel). TRPC1 is currently the strongest candidate component of SOC. Endogenous TRPC1 has been shown to contribute to SOCE (store-operated ...

Septins Arrange F-Actin-Containing Fibers on the Chlamydia trachomatis Inclusion and Are Required for Normal Release of the Inclusion by Extrusion

Chlamydia trachomatis is an obligate intracellular human pathogen that grows inside a membranous, cytosolic vacuole termed an inclusion. Septins are a group of 13 GTP-binding proteins that assemble into oligomeric complexes and that can form higher-order filaments. We report here that the septins SEPT2, -9, -11, and probably -7 form fibrillar structures around the chlamydial inclusion. Colocali...

P to Ca 2 + signaling just got STIMy : an update on STIM 1 activated TRPC channels

Calcium is a ubiquitous signaling molecule, indispensable for cellular metabolism of organisms from unicellular life forms to higher eukaryotes. The biological function of most eukaryotic cells is uniquely regulated by changes in cytosolic calcium, which is largely achieved by the universal phenomenon of store-operated calcium entry (SOCE). The canonical TRPs and Orai channels have been describ...
