چکیده
-Six phenols [2(3)-t-butyl-4-hydroxyanisole (BHA), 2-t-butylphenol, 4-methoxyphenol, 4-methylmercaptophenol, t-butylhydroquinone and 2,6-di-tbutylphenol] previously shown to be inhibitors of benzo(a)pyrene-induced neoplasia, were examined for their ability to induce in vivo changes in hepatic mono-oxygenase and detoxication enzyme activities, and to act as mono-oxygenase inhibitors when added in vitro. (1) Generally it was found that cytochrome P450 levels were depressed, only 2,6-di-t-butylphenol caused a 2-fold induction (2) Mono-oxygenase activities were significantly altered; BHA and 2,6-di-t-butylphenol caused microsomes to show substantial increases in aniline hydroxylase and peroxidase activities. These microsomes, along with 4-methoxyphenol microsomes, also showed a substantial reduction in DNA binding of benzo(a)pyrene (BaP) metabolites relative to metabolism. (3) Detoxication enzymes glutathione S-transferases and epoxide hydratase were readily induced, the order of effectiveness being: BHA -2,6-di-t-butylphenol> 4-methoxyphenol > 2 -t -butylphenol t-butylhydroquinone (4-methylmercaptophenol failed to induce). (4) In vitro ability to inhibit BaP metabolism and DNA-binding ability was: 2,6-di-t-butylphenol , BHA -2-t-butylphenol> t-butylhydroquinone> 4-methylmercaptophenol1>4-methoxyphenol. (5) Ability in vitro to discharge the activated oxygen complex of cytochrome P450 was: 2,6-di-t-butylphenol-,2-t-butylphenol > BHA > t-butlyhydroquinone > 4-methylmercaptophenol > 4-methoxyphenol. The results are consistent with the theory that inhibition of neoplasia is related to inducibility of detoxication enzymes, though alterations in cytochrome P450 could play a significant role in some cases.

In Search of Breast Cancer Culprits: Suspecting the Suspected and the Unsuspected
I would like to welcome breast cancer research community to the first editorial of our newest journal "Breast Cancer: Basic and Clinical Research". In pursuit of breast cancer culprits, we have come a long way since the early 90's when the first breast cancer susceptibility gene BRCA1 was mapped and cloned. In the past few years, several new loci associated with the various degree of breast can...
متن کاملWomen with hereditary breast cancer predispositions should avoid using their smartphones, tablets and laptops at night
Breast cancer is the most common malignancy among women, both in the developed and developing countries. Women with mutations in the BRCA1 and BRCA2 genes have an increased risk of breast and ovarian cancers. Recent studies show that short-wavelength visible light disturb the secretion of melatonin and causes circadian rhythm disruption. We have previously studied the health effects of exposure...
متن کاملA Meta-Analysis on the Relationship between Exposure to ELF-EMFs and the Risk of Female Breast Cancer
OBJECTIVE To comprehensively analyze the relationship between exposure to extremely low frequency electromagnetic fields (ELF-EMFs) and the development of female breast cancer. METHODS Reports of case-control studies published from 1990 to 2010 were analyzed. The quality effect model was chosen to calculate total odds ratio (OR) depending on the data in studies and quality scores. Subgroup an...
متن کاملEffects of 1Hz 100mT electromagnetic field on apoptosis induction and Bax/Bcl-2 expression ratio in breast cancer cells
Introduction: Breast cancer is the most common metastatic malignancy and the second leading cause of death in women. Recently, extremely low-frequency electromagnetic fields (ELF-EMFs) seem to modulate the rate of proliferation and enhance apoptosis and are considered as an emerging approach to cancer therapy. Despite recent success in the electromagnetic fields, the results are still neither d...
متن کامل1Hz 100mT Electromagnetic Field Induces Apoptosis in Breast Cancer Cells Through Up-Regulation of P38 and P21
Introduction: Breast cancer is the most common cause of cancer-related death among women. Recently, extremely low-frequency electromagnetic field (ELF-EMF) has been proposed as a new interfering agent with future therapeutic potentials. Many studies have revealed that cellular processes such as apoptosis in breast cancer are affected by ELF-EMFs. However, more researches are needed to clarify t...
