چکیده

Introduction: Dynamic contrast-enhanced (DCE) MRI is extensively evaluated to assess the potential of biomarkers characterizing tumor microvasculature. Current pharmacokinetic models used with clinically approved small molecular weight contrast agents (CA) yield transfer constants (K) which generally depends on both, capillary permeability and blood flow. In pre-clinical studies, DCE MRI is typically performed on xenograft models in rodents. The non-invasive assessment of an individual arterial input function (AIF) in mouse models remains a challenge [1]. The aim of this study was to develop a technique addressing both issues: First, combining AIF assessment with DCE MRI in a mouse tumor model without the constraint of colocalization of tumor and heart in the same slice. Second, simultaneous measurement of R1 and R2* time courses in the tumor in order to distinguish blood flow and capillary permeability contributions to K .

Automatic Determination of Arterial Input Function for Dynamic Contrast Enhanced MRI in Tumor Assessment

Dynamic Contrast Enhanced MRI (DCE-MRI) is today one of the most popular methods for tumor assessment. Several pharmacokinetic models have been proposed to analyze DCE-MRI. Most of them depend on an accurate arterial input function (AIF). We propose an automatic and versatile method to determine the AIF. The method has two stages, detection and segmentation, incorporating knowledge about artery...

Effect of Bias in Contrast Agent Concentration Measurement on Estimated Pharmacokinetic Parameters in Brain Dynamic Contrast-Enhanced Magnetic Resonance Imaging Studies

Introduction: Pharmacokinetic (PK) modeling of dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) is widely applied in tumor diagnosis and treatment evaluation. Precision analysis of the estimated PK parameters is essential when they are used as a measure for therapy evaluation or treatment planning. In this study, the accuracy of PK parameters in brain DCE...

Simultaneous ΔR1 and ΔR2* Quantification in 5s to Monitor Blood and Tissue Oxygenation with Dynamic (C)O2 Enhanced MRI

The monitoring of respiratory challenges with (C)O2-enhanced MRI is gaining increasing interest for the assessment of tumor oxygenation, vasoreactivity, vessel maturity and function, all being important parameters that decide about optimal treatment strategies [1,2]. In these experiments, a patient’s blood CO2 or O2 level is modulated either by inhalation of (C)O2-enriched air or by breathholds...

Automatic determination of arterial input function for estimating tumor microvessel density with dynamic contrast-enhanced MRI in mice model

Introduction Recently, application of DCE-MRI has gained increasing attention for use in drug development in pre-clinical research using mouse models, especially for assessment of angiogenesis for anti-angiogenic drug development (1). In the mouse, the AIF has been manually measured from a large arterial vessel in the center of the left heart ventricle (2). In this case, when the tumor is far f...

Dynamic Contrast Magnetic Resonance Imaging (DCE-MRI) and Diffusion Weighted MR Imaging (DWI) for Differentiation between Benign and Malignant Salivary Gland Tumors

Background: Salivary gland tumors form nearly 3% of head and neck tumors. Due to their large histological variety and vicinity to facial nerves, pre-operative diagnosis and differentiation of benign and malignant parotid tumors are a major challenge for radiologists. Objective: The majority of these tumors are benign; however, sometimes they tend to transform into a malignant form. Functional M...
