چکیده

Insulin Receptor-related Receptor Messenger Ribonucleic Acid Levels are Upregulated in Enterochromaffin-like Ce11 Pathophysiology. Lisa Anne Ilk. Master of Science, 1 999. Department of Physiology. University of Toronto. The insulin receptor-related receptor (IRR). an orphan member of the insulin receptor family. is selectively localized in the stomach to a subset of neuroendocrine cells. the enterochrornanin-like (ECL) cells. ECL cells are the pivotal regulators of gastric acid secretion. The physiological role of IRR was investigated by analyzing its mRNA levels using semi-quantitative RT-PCR in two States of ECL ce11 pathophysiology in the rat. First. hyperpastrinemia which stimulates ECL ce11 activity and replication was induced by gastric gavage of omepnzole which blocks acid release. Second. a hypogastrinemic state in which ECL ce11 activity was reduced was achieved by fasting. As expected. mRNA levels of the ECL ce11 markers histidine decarboxylase and gastrin receptor increased in hypeqastrinemia and decreased with fasting. IRR mRNA levels also were shown to parallel ihe increase in ECL cell activity in hypergastrinemia. In fasting, however, like the insulin and IGF-1 receptors. IRR mRNA levels increased despite the decrease in ECL ce11 activity. Since IRR mRNA levels were not exclusively conditional on ECL cell activity, they may represent a response to the decreased acid secretion or responses more specific to hypergastrinemia and fasting, for example to increased ECL ce11 activity and loss of ligand, respectively.

Development of real-time reverse transcription polymerase chain reaction assays to quantify insulin-like growth factor receptor and insulin receptor expression in equine tissue.

The insulin-like growth factor system (insulin-like growth factor 1, insulin-like growth factor 2, insulin-like growth factor 1 receptor, insulin-like growth factor 2 receptor and six insulin-like growth factor-binding proteins) and insulin are essential to muscle metabolism and most aspects of male and female reproduction. Insulin-like growth factor and insulin play important roles in the regu...

Effect of fasting on insulin receptor-related receptor messenger ribonucleic acid in rat kidney.

The insulin receptor-related receptor (IRR), a member of the insulin receptor tyrosine kinase family, has structural homology to the insulin receptor (IR) and the IGF-I receptor (IGF-IR). The ligand, gene regulation and biological function of the IRR are not known. Because mRNAs for both the IR and IGF-IR are increased by nutrient restriction, we used RNase protection assays to assess the effec...

ANTISENSE RNA TO THE TYPE I INSULIN-LIKE GROWTH FACTOR RECEPTOR REVERSED THE TRANSFORMED PHENOTYPE OF PC-3 HUMAN PROSTATE CANCER CELL LINE IN VITRO

The insulin-like growth factor I receptor (IGF-IR) plays an essential role in the establishment and maintenance of transformed phenotype. Interference with the IGF-IR pathway by antisense causes reversal of the transformed phenotype in many rodent and human tumor cell lines. We stably transfected the PC-3 human prostate cancer cell line with an IGF-IR antisense RNA expression plasmid. The ...

Expression of melanin-concentrating hormone receptor messenger ribonucleic acid in tumor tissues of pheochromocytoma, ganglioneuroblastoma, and neuroblastoma.

Expression of melanin-concentrating hormone (MCH) receptor messenger ribonucleic acid (mRNA) was studied by RT-PCR and Northern blot analysis in human brain; pituitary; adrenal glands; tumor tissues of adrenal tumors, ganglioneuroblastomas, and neuroblastomas; and various cultured tumor cell lines. RT-PCR analysis showed that MCH receptor mRNA was widely expressed in brain tissues, pituitary, n...

The effect of human wharton’s jelly-derived mesenchymal stem cells on MC4R, NPY, and LEPR gene expression levels in rats with streptozotocin-induced diabetes

Objective(s): Type 1 diabetes (T1D) is an autoimmune disease resulting from inflammatory destruction of islets β-cells. Nowadays, progress in cell therapy, especially mesenchymal stem cells (MSCs) proposes numerous potential remedies for T1D. We aimed to investigate the combination therapeutic effect of these cells with insulin and metformin on neuropeptide Y, melanoco...
