چکیده

L-H represents a lipid, generally a polyunsaturated fatty acid moiety (PUFA). (See Figure 1 legend for an explanation of the abbreviations used to denote the various chemical species associated with lipid peroxidation). The rate of the propagation event is governed by the various carbon-hydrogen bond dissociation energies along the lipid chain (Cosgrove et al., 1987). The weakest carbonhydrogen bonds are those of the bis-allylic methylene positions, which have carbon-hydrogen bond energies of approximately 75 kcal/mol compared to 101 kcal/mol for typical alkyl C-H bonds (Figure 2). In homogeneous solution the oxidizability of unsaturated lipids increases linearly with the number of bis-allylic positions in the lipid chain (Cosgrove et al., 1987). However, the rate of cellular lipid peroxidation has been shown to increase exponentially with the number of bisallylic positions contained in cellular lipids (Wagner et al., 1994).

Acidic pH amplifies iron-mediated lipid peroxidation in cells.

The goal of our study was to investigate the mechanism by which changes in extracellular pH influence lipid peroxidation processes. Ferrous iron can react with hydroperoxides, via a Fenton-type reaction, to initiate free radical chain processes. Iron is more soluble at lower pH values, therefore we hypothesized that decreasing the environmental pH would lead to increased iron-mediated lipid per...

Lipid hydroperoxide generation, turnover, and effector action in biological systems.

Lipid peroxidation is a well known example of oxidative damage in cell membranes, lipoproteins, and other lipid-containing structures. Peroxidative modification of unsaturated phospholipids, glycolipids, and cholesterol can occur in reactions triggered by i) free radical species such as oxyl radicals, peroxyl radicals, and hydroxyl radicals derived from iron-mediated reduction of hydrogen perox...

Ultraviolet A-induced peroxidation of phosphatidylcholine in unilamellar liposomes.

Unilamellar liposomes of 1-palmitoyl-2-linoleoyl-3-sn-phosphatidylcholine (PLPC) were exposed to ultraviolet A (UVA) light at 37 degrees C. The UVA-induced PLPC peroxidation was inhibited by free radical scavengers, while singlet oxygen quenchers showed slight inhibition. When pre-existing hydroperoxides were reduced, UVA irradiation could not initiate the lipid peroxidation. The UVA-induced re...

Singlet oxygen production associated with enzyme-catalyzed lipid peroxidation in liver microsomes.

Evidence for the formation of singlet oxygen during the oxidation of NADPH by liver microsomes is presented. The evidence is based primarily on the enzyme-dependent formation of dibenzoylethylene from diphenylfuran, a reaction which is specific for singlet oxygen. The apparent formation of singlet oxygen is coupled to the occurrence of peroxidation of microsomal lipid, a phenomenon known to be ...

Simulation of free radical reactions in biology and medicine: a new two-compartment kinetic model of intracellular lipid peroxidation.

To explore mechanisms of free radical reactions leading to intracellular lipid peroxidation in living systems, we developed a computational model of up to 109 simultaneous enzymatic and free radical reactions thought to be involved in the initiation, propagation, and termination of membrane lipid peroxidation. Rate constants for the various reactions were obtained from the published literature....
