The Agilent InfinityLab 2D-LC Solution with Active Solvent Modulation Achieving Improved Resolution and Sensitivity for Challenging Combinations of Separation Conditions

چکیده

A major challenge during method development in two-dimensional liquid chromatography (2D-LC) can be the incompatibility between the first (1D) and second dimension (2D) solvent systems. For challenging combinations of separation systems, volume overload effects such as breakthrough or broad and distorted peaks in the 2D can be observed. Active solvent modulation (ASM) provides a valve-based approach to address this problem, leading to improved resolution and sensitivity in the 2D. This Technical Overview explains the principle of ASM, and demonstrates its effect in the combination of two reversed-phase (RP) LC separations and a combination of hydrophilic interaction liquid chromatography (HILIC) and RPLC. High-Resolution Sampling 2D-LC Analysis of Mangosteen Pericarp Xanthones Using the Agilent 1290 Infinity II 2D-LC Solution

A high-resolution UHPLC analysis of xanthones present in the extracts of the pericarp of mangosteen (Garcinia mangostana L.) was carried out on an Agilent 1290 Infinity II LC. Excellent resolution and peak capacity (300 calculated at half height) were obtained within a total analysis time, including column reconditioning, of 16.5 minutes. The main xanthone (α-mangostin) was quantified in two ex...

Detection of impurities by heart cutting using the Agilent 1290 Infi nity 2 D - LC Solution Application

This Application Note demonstrates the capability of the Agilent 1290 Infi nity 2D-LC Solution for heart-cutting applications. As an example, a minor impurity is resolved by heart cutting from a co-eluting main compound. The minor impurity, which is isolated on a second-dimension column after heart cutting, is verifi ed by additional spiking and statistical evaluation of the separation performa...
