چکیده

In the cultured HEK model, we observed that mustard stimulates protease activity, and the epidermaldermal attachment protein laminin-5 is a substrate. Addition of serine protease inhibitors (50 μM ICD 2812 or 1 mM phenyl methyl sulphonyl fluoride (PMSF)), the metalloprotease inhibitor 1, 10phenanthroline (1 mM), or the general caspase inhibitor Z-VAD-FMK (benzyl oxycarbonyl-Val-Ala-Asp (o-methyl-fluromethylketone), 10 μM) to cells prior to mustard decreased the protease bands (zymography) and laminin-5 degradation (Western blotting, immuno-fluorescence). In conclusion, our results indicate that (a) mustard stimulates multiple proteases in the skin, and (b) protease inhibitors are prospective vesicant countermeasures.

Antivesicant Strategies Based on Dna Repair and Apoptosis

DNA is a major cellular target of the vesicant chemical warfare agent sulfur mustard (SM, bis-(2-chloroethyl) sulfide). Others and we have proposed a possible role of apoptosis in SM vesication. Our results suggest that in SM-exposed human epidermal keratinocytes (HEK), DNA damage, DNA repair, and apoptosis may be interdependent. In HEK, SM causes cell death accompanied by caspase-3 activation ...

Sulfur mustard-stimulated proteases and their inhibitors in a cultured normal human epidermal keratinocytes model: A potential approach for anti-vesicant drug development

Protease stimulation in cultured normal human epidermal keratinocytes (NHEK) due to sulfur mustard (SM) exposure is well documented. However, the specific protease(s) stimulated by SM and the protease substrates remain to be determined. In this study, we observed that SM stimulates several proteases and the epidermal-dermal attachment protein laminin-5 is one of the substrates. We propose that ...

Molecular Docking Based on Virtual Screening, Molecular Dynamics and Atoms in Molecules Studies to Identify the Potential Human Epidermal Receptor 2 Intracellular Domain Inhibitors

Human epidermal growth factor receptor 2 (HER2) is a member of the epidermal growth factor receptor family having tyrosine kinase activity. Overexpression of HER2 usually causes malignant transformation of cells and is responsible for the breast cancer. In this work, the virtual screening, molecular docking, quantum mechanics and molecular dynamics methods were employed to study protein–ligand ...

The Role of Energy Metabolism in Cutaneous Sulfur Mustard Injury

Over the past 17 years, our research efforts have focused on systematic studies of the effects of sulfur mustard (2,2'-dichlorodiethyl sulfide, HD) on energy metabolism in human epidermal keratinocytes (HEK). Our objectives are to define mechanisms of HD-induced metabolic injury, determine their role in the cutaneous pathology observed and provide mechanistic information that can be used in dev...
