چکیده

Streptococcus agalactiae, or GBS, is an important opportunistic pathogen which causes pneumonia, sepsis and meningitis in neonates and severe diseases in immunocompromised adults. We have performed comparative genomics of prevalent GBS serotypes of Indian origin i.e., Ia, III, V and VII. Pili proteins were commonly found upregulated and their expression was studied by using antiserum for GBS80 (backbone protein of pilus island–I (PI1), GBS67 (ancillary protein of PI-2a) and SAN1518 (backbone protein of PI2b) by whole cell and Western blot analysis. To check the role of pili proteins in adherence and invasion, inhibition assay was performed. Comparative immunoblotting experiments revealed that pili proteins expression does not differ in geographically different selected serotypes, Ia and V of India and USA. In case of A549 we found that GBS VII invasion and adherence was inhibited by pili protein specific antiserum SAN1518 significantly (P=<0.001) by 88.5 and 91 % respectively. We found that mutant strains, deficient in the pilus proteins (Δgbs80, and Δsan1518 exhibit a significant decrease in adherence in case of type Ia, III, VII. We have found 95 % reduction in invasion when ∆SAN1518 was used in case of A549 cells. As the pili proteins were identified previously as vaccine candidates against GBS serotypes of developed countries and we also found their role in the attachment and invasion of GBS of Indian origin thus in turn present work supports the idea of making a more effective pili protein based vaccine which can be used universally.

Comparative Molecular and Microbiologic Diagnosis of Vaginal Colonization by Group B Streptococcus in Pregnant Women during Labor

Objective(s) Rapid tests for detection of Streptococcus agalactiae or Group B Streptococci (GBS) at the onset of labor are needed to permit early intrapartum antibiotic prophylaxis. This study aimed to evaluate the PCR assays targeting the 16S ribosomal RNA gene (16S rDNA) for detection of the GBS in comparison with a specific culture method. Materials and Methods Two swabs were used to obta...

Maternal Carriage and Neonatal Colonization of Streptococcus agalactiae in Tabriz, Northwest Iran

Group B Streptococcus (GBS) is a major cause of severe systemic and local infections in newborns. This study was performed to evaluate the presence of GBS in pregnant women and their newborns in Tabriz, Iran. Vaginal specimens were collected from 965 women who were candidate for vaginal delivery for bacteriological study of group B streptococci. Several samples from various sites of the newborn...

Regulation and function of pilus island 1 in group B streptococcus.

Group B streptococcus (GBS) pili may enhance colonization and infection by mediating bacterial adhesion to host cells, invasion across endothelial and epithelial barriers, and resistance to bacterial ingestion and killing by host phagocytes. However, it remains unclear how pilus expression is regulated and how modulation of pilus production affects GBS interactions with the human host. We inves...

Development of an indirect ELISA for bovine mastitis using Sip protein of Streptococcus agalactiae

The sip gene encoding for a conserved highly immunogenic surface protein of Streptococcus agalactiae was amplified using polymerase chain reaction (PCR) and subcloned into prokaryotic expression vector pET32a (+) and expressed as a recombinant protein in E. coli BL21 (DE3). An indirect enzyme linked immunosorbent assay (ELISA) was developed using the purified Sip protein as a coating antigen, w...
