The mitochondrial compartment.

چکیده

Mitochondria are vital organelles that perform a variety of fundamental functions ranging from the synthesis of ATP through to being intimately involved in programmed cell death. Comprised of at least six compartments: outer membrane, inner boundary membrane, intermembrane space, cristal membranes, intracristal space, and matrix, mitochondria have a complex, dynamic internal structure. This internal dynamism is reflected in the pleomorphy and motility of mitochondria. Mitochondria contain their own DNA (mtDNA), encoding a small number of vital genes, but this role as a genetic vault is not compatible with the role of mitochondria in bioenergetics since electron transport results in the generation of reactive oxygen species (ROS) that induce lesions in the mtDNA. It is hypothesized that ROS shape the morphological organization of the higher plant cell mitochondrial population into a discontinuous whole, and that ROS are a selective pressure affecting the organization of the mitochondrial genome. This review describes how inter- and intra-mitochondrial compartmentalization underpins the biology of this complex organelle.

Topological and functional organization of the mitochondrion.

The morphological and biochemical properties of isolated mitochondrial inner and outer membranes are summarized and discussed in relation to the functional organization of the intact mitochondrion. The enzymatic composition of the mitochondrial inner compartment is compared to over-all mitochondrial function. The role of the mitochondrial outer compartment is discussed with reference to both th...

Mitochondrial turnover in animal cells. Half-lives of mitochondria and mitochondrial subfractions of rat liver based on [14C]bicarbonate incorporation.

The half-lives of proteins of mitochondria and a well characterized set of mitochondrial subfractions have been estimated using two methods based on incorporation of a minimally reutilized amino acid precursor, [‘‘Clbicarbonate. The apparent average rate constants of protein degradation were measured on rat liver homogenate, mitochondria, and distinct outer compartment, inner membrane, and matr...

Mitochondrial Disulfide Relay: Redox-regulated Protein Import

99% of all mitochondrial proteins are synthesized in the cytosol, from where they are imported into mitochondria. In contrast to matrix proteins, many proteins of the intermembrane space (IMS) lack presequences and are imported in an oxidation-driven reaction by the mitochondrial disulfide relay. Incoming polypeptides are recognized and oxidized by the IMSlocated receptor Mia40. Reoxidation of ...

Constriction of the mitochondrial inner compartment is a priming event for mitochondrial division

Mitochondrial division is critical for the maintenance and regulation of mitochondrial function, quality and distribution. This process is controlled by cytosolic actin-based constriction machinery and dynamin-related protein 1 (Drp1) on mitochondrial outer membrane (OMM). Although mitochondrial physiology, including oxidative phosphorylation, is also important for efficient mitochondrial divis...

Compartment Syndrome of the Calf Due to Nicolau Syndrome

We report a case of Nicolau syndrome in a 15 months old girl following of an intramuscular injection of penicillin 6.3.3 in her left buttock. This case is unique because she developed compartment syndrome in her left calf far from her injection site. Her toe’s tips gangrened in the course of her ailment. We hypothesized that the compartment syndrome might be produced by a probable intra-arteri...

Compartment syndrome as a result of incorrect treatment

Compartment syndrome is a rare vascular disorder and an orthopedic emergency caused by high intramuscular pressure following bone fractures and some other etiologies. It mostly involves extremities, but can affect other parts of the body. The syndrome is diagnosed based on extensively varying signs and symptoms including feeling pain, pallor, pulselessness, and some other signs among patients. ...
