Effect of chemical modification of dihydrouridine in yeast transfer ribonucleic acid on amino acid acceptor activity and ribosomal binding.

چکیده
Dihydrouridine residues in bakers’ yeast transfer RNA were reductively cleaved with sodium borohydride. The amino acid acceptor activity of three species of transfer RNA and the ribosoma binding of one species of transfer RNA were determined for these modified transfer RNAs. The data indicate that the reductive cleavage of the dihydrouridine residues in these transfer RNAs has no effect upon their ability to accept amino acids or to bind to ribosomes.

tertiary structure of the molecule. These seem to be in the region of the ‘hinge’ between dihydrouridine and the ribosylthymine-pseudouridine loop. Further, the n.m.r. studies allow one to propose a detailed structural model for the anticodon loop (Ts’o et al., 1975). The problem of the specificity of aminoacylation has been approached by equilibrium and kinetic studies. A striking feature of t...
متن کاملAction of spleen exonuclease on transfer ribonucleic acid.
The hydrolytic action of spleen exonuclease on serine transfer RNA has been investigated. It was found that two different mechanisms of attack obtain: (a) processive degradation and (b) random cleavage. These two diiferent modes of action are determined by the presence or the absence of the terminal phosphate group at the 5’ terminus of transfer RNA. The loss of several important biological act...
متن کاملEffect of Amino Acid Substitutions on Biological Activity of Antimicrobial Peptide: Design, Recombinant Production, and Biological Activity
Recently, antimicrobial peptides have been introduced as potent antibiotics with a wide rangeof antimicrobial activities. They have also exhibited other biological activities, including antiinflammatory, growth stimulating, and anti-cancer activities. In this study, an analog of MagaininII was designed and produced as a recombinant fusion protein. The designed sequence containe...

متن کاملFractionation of Yeast Amino Acid-Acceptor Ribonucleic Acids by Countercurrent Distribution*?
Considerable interest has developed in recent years in the separation of the amino acid-specific “soluble’‘-fraction ribonucleic acids because of the probable role of these RNAs in protein biosynthesis (l-5) and because the elucidation of the structures of the different amino acid-specific RNAs may lead to an understanding of the mechanism of information transfer by nucleic acids. Preliminary s...
