چکیده

The survival of Xanthomonas campestris pv. musacearum (Xcm), the cause of banana bacterial wilt disease in soil and infected banana debris was investigated. Viable cells of the pathogen were estimated on culture medium. The survival of the pathogen in soil was influenced by soil moisture and soil treatment. The survival period of Xcm was reduced 3 times when soil moisture content was reduced from 28% to 14%. Soil treatment impacted on the survival of Xcm, with populations declining rapidly in non-sterile soil than in sterile soil. No viable cells of the pathogen were recovered from non-sterile soil after 20 days under both high and low soil moisture contents. Viable pathogen cells survived for slightly a longer period in soil in the field than non-sterile soil under controlled conditions. In sterile soil the pathogen cells persisted for up to 90 and 45 days under high and low moisture soil respectively. Populations of Xcm in debris in the field declined rapidly, with no pathogens cells recovered after 21 days but the viable cells remained stable in the laboratory throughout the 90 days of sampling. Results indicate that Xcm has limited ability to survive saprophytically in soil and plant debris in presence of other competing microoganisms. This implies that bananas can be replanted in fields where the crop was previously destroyed by Xcm in a relatively short period of time.

Role of Soil, Crop Debris, and a Plant Pathogen in Salmonella enterica Contamination of Tomato Plants

BACKGROUND In the U.S., tomatoes have become the most implicated vehicle for produce-associated Salmonellosis with 12 outbreaks since 1998. Although unconfirmed, trace backs suggest pre-harvest contamination with Salmonella enterica. Routes of tomato crop contamination by S. enterica in the absence of direct artificial inoculation have not been investigated. METHODOLOGY/PRINCIPAL FINDINGS Thi...

Use of bioluminescence for detection of genetically engineered microorganisms released into the environment.

The persistence and movement of strain JS414 of Xanthomonas campestris pv. campestris, which was genetically engineered to bioluminesce, were monitored during a limited field introduction. Bioluminescence and traditional dilution plate counts were determined. Strain JS414 was applied to cabbage plants and surrounding soil by mist inoculation, by wound inoculation, by scattering infested debris ...

Genetically engineered bananas resistant to Xanthomonas wilt disease and nematodes

Banana is an important staple food crop feeding more than 100 million Africans, but is subject to severe productivity constraints due to a range of pests and diseases. Banana Xanthomonas wilt caused by Xanthomonas campestris pv. musacearum is capable of entirely destroying a plantation while nematodes can cause losses up to 50% and increase susceptibility to other pests and diseases. Developmen...

Genome-Wide Sequencing Reveals Two Major Sub-Lineages in the Genetically Monomorphic Pathogen Xanthomonas Campestris Pathovar Musacearum

The bacterium Xanthomonas campestris pathovar musacearum (Xcm) is the causal agent of banana Xanthomonas wilt (BXW). This disease has devastated economies based on banana and plantain crops (Musa species) in East Africa. Here we use genome-wide sequencing to discover a set of single-nucleotide polymorphisms (SNPs) among East African isolates of Xcm. These SNPs have potential as molecular marker...

Draft Genome Sequences of Xanthomonas sacchari and Two Banana-Associated Xanthomonads Reveal Insights into the Xanthomonas Group 1 Clade

We present draft genome sequences for three strains of Xanthomonas species, each of which was associated with banana plants (Musa species) but is not closely related to the previously sequenced banana-pathogen Xanthomonas campestris pathovar musacearum. Strain NCPPB4393 had been deposited as Xanthomonas campestris pathovar musacearum but in fact falls within the species Xanthomonas sacchari. St...
