چکیده

A direct relation between gene dosage and amount of the corresponding protein is generally assumed to exist though it has been investigated in a limited number of cases. Yeasts seem to be an ideal material for this type of investigation as they can be easily obtained in different degrees of ploidy. Macromolecular components of yeast cells in polyploid series have been already determined (OGUR et al. 1952) and the relationships between gene dosage and enzyme production have been analysed for galactokinase (NELSON and DOUGLAS 1963) and iso-lcytochrome (SJXNIMSKI et al. 1965). Data reported here will deal with the con-' struction of a polyploid series of Saccharomyces cerevisiae (haploid, diploids and tetraploids) , the determination of DNA and soluble protein content on a per cell basis, and the relation of tryptophan synthetase2 (TST) activity to ploidy and gene dosage.

Inhibitory Effect of Supernatant and Lysate of Saccharomyces cerevisiae on Expression of exoA Gene of Pseudomonas aeruginosa

Background and Aim: Pseudomonas aeruginosa is an important ubiquitous and especially common pathogen in the hospital. Exotoxin A that encoded by exoA gene has a role in pathogenesis of this bacterium. Today, probiotics are widely used in the treatment and prevention of diseases. The present study aimed to study the Saccharomyces cerevisiae S3 effect on the expression of exoA gene. Materials an...

Two mechanisms of allelic complementation among tryptophan synthetase mutants of Saccharomyces cerevisiae.

Two different types of allelic complementation were observed in tryptophan synthetase mutants of the yeast Saccharomyces cerevisiae. Each type is associated with a different mechanism for the enzymatic conversion of indole-3-glycerol phosphate (InGP) to tryptophan. Mechanism I is utilized by a hybrid tryptophan synthetase that resembles, but is not identical with, the wild-type enzyme. Mechanis...

Identification of a Glutaminyl - tRNA Synthetase Mutation in Saccharomyces cerevisiae AARON

Saccharomyces cerevisiae glutaminyl-tRNA synthetase mutants were isolated through systematic screening of tight Glnderivatives of a leaky glutamine auxotroph. These mutations define a single nuclear gene, GLN4. The gln4-1 mutation is specific for Gln-tRNA synthetase and shows a dosage effect in heterozygous diploids. The wild-type Gln-tRNA synthetase exhibits a Km for glutamine of 25 ,uM; the g...
