چکیده

An assay technique, homogeneous enzyme immunoassay, is described for the quantitative determination of morphine derivatives in biological fluids. An enzyme is labeled with morphine. When the enzymelabeled morphine is bound by antimorphine antibodies, the enzyme is rendered inactive. Free morphine competes with enzyme-morphine for antibody binding sites preventing inhibition of enzyme activity. Enzyme activity is thus directly related to the concentration of free morphine.

Development and GC-MS validation of a highly sensitive recombinant G6PDH-based homogeneous immunoassay for the detection of buprenorphine and norbuprenorphine in urine.

Buprenorphine is now increasingly prescribed as an alternative to methadone for the treatment of heroin addiction. Because of its potency (dosage usages from 0.2 mg to 8 mg), the drug concentrations in body fluids are normally very low. Here, we report the first recombinant glucose-6-phosphate dehydrogenase (G6PDH)-based homogeneous immunoassay (EMIT-type assay) for free buprenorphine and free ...

Urinary buprenorphine concentrations in patients treated with suboxone as determined by liquid chromatography-mass spectrometry and CEDIA immunoassay.

We report on the utility of urine total buprenorphine, total norbuprenorphine, and creatinine concentrations in patients treated with Suboxone (a formulation containing buprenorphine and naloxone), used increasingly for the maintenance or detoxification of patients dependent on opiates such as heroin or oxycodone. Patients received 8-24 mg/day buprenorphine. Two-hundred sixteen urine samples fr...

Clinical issues associated with urine testing of substances of abuse.

Several factors may affect the validity and outcome of urine testing for abused drugs such as amphetamines, barbiturates, benzodiazepines, cannabinoids, cocaine, ethanol, opiates, and phencyclidine. Urine is used for large-scale testing because acquisition of the sample is noninvasive and because most abused drugs can be detected in urine for a reasonable duration after ingestion. Urine testing...

Laboratory analysis of remotely collected oral fluid specimens for opiates by immunoassay.

The performance characteristics of a method for detecting opiates (morphine, codeine, heroin, and 6-acetylmorphine [6-AM]) in oral fluid specimens were examined and compared with methods for urine specimens. The oral fluid was easily obtained using a simple device that collects between 1 and 1.5 mL of fluid for laboratory analysis. Simultaneously collected specimens from 60 known opiate abusers...

Comparison of the EMIT (enzyme multiplied immunoassay technique) opiate assay and a gas-chromatographic--mass-spectrometric determination of morphine and codeine in urine.

We examined 124 urine samples with the EMIT opiate assay kit and with a gas-chromatographic-mass-spectrometricdetermination (I) for morphine and codeine. With a cut-off value between positive and negative resultsat0.5 mg (morphine equivalents)per liter for both methods, the EMIT assay gave 4.0% falsepositivesand 5.6% false negativeswhen compared with I. Lowering of the cut-off value for Ito 0.1...
