Differential binding of antiphospholipid autoantibody in normal and autoimmune sera to prothrombin/phospholipid antigen complex.

چکیده
Oosting et al's' recent report on various phospholipid/protein antigen complexes (involving prothrombin, protein S, or protein C) recognized by antiphospholipid antibodies (aPL) has set the stage for other aPL investigators to redefine and evaluate the binding specificity of aPL. The nature of the neoepitopes induced by protein/phospholipid interactions is still unresolved. Firstly, the antigenicity of the phospholipid may be changed' or, alternatively, cryptic epitopes may be expressed on the protein cofactors when associated with phospholipid^.^ Shared epitopes formed between the protein and phospholipid could also be potentially involved in the binding reactivity of aPL. Naturally occumng autoantibodies, including aPL, are increasingly found as a common feature in normal human sera (NHS)?,5 A proportion of natural aPL are "masked" in NHS, but can be shown and detected in enzyme-linked immunosorbent assay (ELISA) by prior heat-inactivation of the sera at 56°C for 30 minu t e ~ . ~ These natural aPL may, under certain dysfunctions in immunoregulation, become pathogenetic? The aPL serum cofactor, 0'glycoprotein I (O2-GPI) has been shown to affect the binding of natural aPL differently from its enhancing role for autoimmune aPL. In phospholipid ELISA, D2-GPI, when complexed with the phospholipid, reduced the IgG aPL binding in NHS.6.' We have now observed a similar differential effect of purified prothrombin on aPL binding. In cardiolipin phospholipid ELISA, the addition of prothrombin decreased the natural IgG aPL binding5 (Table l). There was negligible binding of natural aPL to prothrombin alone. This indicates that the molecular presentation of prothrombin/phospholipid complex is altered, with resultant greater IgG aPL binding in autoimmune sera, but is unfavorable for natural IgG aPL binding. New clinical and laboratory finding in anti-phospholipid syndromes
Antiphospholipid syndrome (APS) is a systemic autoimmune disease with the appearance of anti-phospholipid antibody (ACLA), Anti β2GPI and lupus anticoagulant (LAC) in serum. The antigen-antibody reaction on cell surface leads to thrombosis, miscarriage and inflammation. Clinical findings are diverse and include thrombosis in veins and arteries, pregnancy loss, thrombocytopenia, neurological and...
متن کاملPrevalence of different anti-phospholipid antibodies in systemic lupus erythematosus and their relationship with the antiphospholipid syndrome.
The antiphospholipid syndrome is a complex disorder (1 ) that is related to a set of unusual and fascinating antibodies. Recent research is defining these antibodies and their relationship to features of the syndrome. The antiphospholipid syndrome is defined by the association of arterial and/or venous thrombosis, recurrent miscarriages, and anti-phospholipid (aPL) antibodies (1 ). The syndrome...
متن کاملAntiphospholipid antibodies accelerate plasma coagulation by inhibiting annexin-V binding to phospholipids: a "lupus procoagulant" phenomenon.
The antiphospholipid syndrome is a thrombophilic condition marked by antibodies that recognize anionic phospholipid-protein cofactor complexes. We recently reported that exposure to IgG fractions from antiphospholipid patients reduces the level of annexin-V, a phospholipid-binding anticoagulant protein, on cultured trophoblasts and endothelial cells and accelerates coagulation of plasma exposed...
متن کاملSynthesis of Phospholipid-Protein Conjugates as New Antigens for Autoimmune Antibodies.
Copper(I)-catalyzed azide-alkyne cycloaddition, or CuAAC click chemistry, is an efficient method for bioconjugation aiming at chemical and biological applications. Herein, we demonstrate how the CuAAC method can provide novel phospholipid-protein conjugates with a high potential for the diagnostics and therapy of autoimmune conditions. In doing this, we, for the first time, covalently bind via ...

When pooled immunoglobulin G (IgG) from patients with multiple sclerosis (MS) was used to probe a human fetal spinal cord λgt 11 cDNA library, the IgG was found to bind to a predicted epitope of human mitochondrial ND4 sequence. To investigate the involvement of the ND4 as an autoantigen in MS, we determined the presence of specific antibody in the serum of MS patients and serum samples of...
