Stability of Freeze-dried Inactivated Japanese Encephalitis Vaccine under Different Experimental Conditions
نویسندگان
چکیده
Stability of three batches of freeze dried inactivated mouse brain Japanese encephalitis vaccine, was evaluated in terms of percentage drop or otherwise in potency from their respective initial titers, after exposure to different experimental conditions for 4 -12 weeks. The potency was assayed by plaque reduction neutralization assay (PRNT) using chick embryo fibroblasts cells. The vaccine has been found to be stable i.e. retain potency (mean drop 3.99%) till 12 weeks when exposed at 25 0 C ± 2 0 C and 80-90% relative humidity (RH). At 37 0 C ± 1 0 C and when kept in vertical position, the vaccine showed an average drop of 10.42% in potency after 12 weeks, while it was 11.0% and 10.42% in horizontal positions and inverted position respectively. Exposure at higher temperature (45 0 C + 1 0 C and 80-90% RH) led to deterioration of potency (25.82%) after 8 weeks. A maximum average drop of 7.66% in potency was observed when vaccine was exposed to sunlight compared to 4.29% in case of artificial light after 12 weeks indicating that the JE vaccine should not be exposed to direct light. Thus, even if this vaccine gets exposed to higher temperature for a short duration, it appears that FD JE vaccine may be potent enough for effective seroconversion in the

Phase III clinical trials comparing the immunogenicity and safety of the vero cell-derived Japanese encephalitis vaccine Encevac with those of mouse brain-derived vaccine by using the Beijing-1 strain.
The immunogenicity and safety of an inactivated cell culture Japanese encephalitis vaccine (CC-JEV) were compared with those of an inactivated mouse brain-derived Japanese encephalitis vaccine (MB-JEV) in phase III clinical multicenter trials conducted in children. The vaccines contain the same Japanese encephalitis virus strain, the Beijing-1 strain. Two independent clinical trials (trials 1 a...

A stable B.C.G. vaccine, prepared and distributed in the freeze-dried form, is now accepted for use in many parts of the world. Recent trials (Medical Research Council, 1958, 1960; Griffiths and Gaisford, 1956) have confirmed that the freeze-dried vaccine induces a high degree of tuberculin sensitivity without untoward local reactions. From its stability the freeze-dried vaccine has undoubted a...

Five different subtypes of foot and mouth disease (FMD) virus propagated in BHK21 C13 monolayer cell cultures, namely 05, A5/!0, A22, C¡ and Asia 1/1, were used to study the stability of live virus during freeze-drying and subsequent storage. Binary ethyleneimine-inactivated FMD virus vaccines adjuvanted with purified saponin, "Quil-A", without aluminium hydroxide gel, were examined before and ...

One of the advantages of dry influenza vaccines over conventional liquid influenza vaccines is that they can be used for alternative routes of administration. Previous studies showed that spray freeze-drying is an excellent technique to prepare vaccine containing powders for pulmonary delivery (J.P. Amorij, V. Saluja, A.H. Petersen, W.L.J. Hinrichs, A. Huckriede, H.W. Frijlink, Pulmonary delive...
متن کاملSafety and immunogenicity of a freeze-dried, Vero cell culture-derived, inactivated Japanese encephalitis vaccine (KD-287, ENCEVAC®) versus a mouse brain-derived inactivated Japanese encephalitis vaccine in children: a phase III, multicenter, double-blinded, randomized trial
BACKGROUND Although mouse brain-derived, inactivated Japanese encephalitis vaccines (JE-MBs) have been successfully used for a long time, potential rare neurological complications have prompted the development of a Vero cell culture-derived inactivated vaccine (JE-VC). In a phase III clinical study, we aimed to compare the safety and immunogenicity of a JE-VC, KD-287 with a JE-MB, JEV-GCC, in c...
