Recent aspects of the regulation of intestinal mucus secretion.

چکیده
L’kpithklium spkcialisi qui borde la muqueuse intestinale n’est pas seulement une barrikre passive entre la lumitxe et le milieu interieur; c’est un systkme complexe qui participe activement ?I la dkfense de la muqueuse. En particulier, les fonctions de transport (IgA) , de skcrktion vectorielle d’klectrolytes (Cl-) et de macromolkcules (mucines) constituent la premikre ligne de defense de cet Cpithklium. Ces fonctions de skcrbtion sont rCgulCes par des agents neuroendocrines et des mkdiateurs de l’inflammation et de I’immunitC. Le dkveloppement de modkles ‘in vitro’ permettant d’ktudier le mode d’action de ces mkdiateurs sur les fonctions skcrktoires intestinales est au coeur de la recherche en gastroentkrologie. Les cultures dites organotypiques fondkes sur le maintien in vitro de fragments de muqueuse intestinale et les prkparations de cellules epithkliales isolkes ont de skrieuses limitations notamment en termes de viabilitk cellulaire. C’est pourquoi les recherches rkcentes se sont orientkes vers le dkveloppement de lignkes cellulaires diffkrencikes et homogknes, issues de cancers coliques humains, qui maintiennent in vitro les fonctions diffkrencikes qui sont celles des colonocytes in vivo. Nous prksentons ici l’ktablissement et l’exploitation du modkle HT29-Cl.l6E, issu de notre laboratoire, pour l’ktude des rkcepteurs, de la signalisation intracellulaire et enfin des mkcanismes distaux de l’exocytose des cellules 2 mucus de l’intestin.
