NF-kB controls the global pro-inflammatory response in endothelial cells: evidence for the regulation of a pro-atherogenic program

چکیده
Activation of the transcription factor NF-kB is critical for the tumor necrosis factor-a (TNF-a)-induced inflammatory response. Here we report the complete gene expression profile from activated microvascular endothelial cells emphasizing the direct contribution of the NF-kB pathway. Human microvascular endothelial cell line-1 (HMEC-1) cells were modified to express dominant interfering mutants of the IKK/NF-kB signaling module and expression profiles were determined. Our results provide compelling evidence for the virtually absolute dependence of TNF-a-regulated genes on NF-kB. A constitutively active IKK2 was sufficient for maximal induction of most target genes, whereas a transdominant IkBa suppressed gene expression. Several genes with a critical role in atherogenesis were identified. The endothelial lipase (EL) gene, a key enzyme involved in lipoprotein metabolism, was investigated more in detail. Binding sites interacting with NF-kB in vitro and in vivo were identified and co-transfection experiments demonstrated the direct regulation of the EL promoter by NF-kB. We conclude that targeting the IKK/NF-kB pathway or specific genes downstream may be effective for the control or prevention of chronic inflammatory diseases such as

متن کاملThe effect of down-regulation of CCL5 on lipopolysaccharide-induced WI-38 fibroblast injury: a potential role for infantile pneumonia
Objective(s): Aberrant expression of CCL5 has been found in several kinds of inflammatory diseases, and the roles of CCL5 in these diseases have also been reported. However, the role of CCL5 in infantile pneumonia is still unclear. Thus, the function and acting mechanism of CCL5 in the in vitro model of infantile pneumonia were researched in this study. Materials and Methods: Human fetal lung f...
متن کاملHEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Overexpression of A1, an NF-kB–Inducible Anti-Apoptotic Bcl Gene, Inhibits Endothelial Cell Activation
A1 is an anti-apoptotic bcl gene that is expressed in endothelial cells (EC) in response to pro-inflammatory stimuli. We show that in addition to protecting EC from apoptosis, A1 inhibits EC activation and its associated expression of proinflammatory proteins by inhibiting the transcription factor nuclear factor (NF)-kB. This new anti-inflammatory function gives a broader dimension to the prote...

متن کاملThe Effect of Combining Aerobic Exercise and Royal Jelly on the Expression of Pro-Inflammatory Cytokines in Liver Tissue of Obese Rats
Background: Inflammation plays a critical role in the promotion of hepatocyte damage and liver fibrosis. In recent years the protective role of exercise and royal jelly (RJ), has been suggested for metabolic and inflammatory disorders. In this study, we aimed to evaluate the effects of aerobic training (AT) and RJ on pro-inflammatory markers in a rat's model of a high-fat diet (HFD). Methods: ...
