چکیده

The storage triacylglycerols of nasturtium (Tropaeolum majus) seeds are composed principally of cis-11-eicosenoate and cis-13-docosenoate. To investigate the biosynthesis of these C(20) and C(22) fatty acids, developing seed tissue was incubated with various (14)C-labeled precursors. Incubation with [1-(14)C]acetate produced primarily cis-11-[1-(14)C]eicosenoate and cis-13-[1,3-(14)C]docosenoate in the triacylglycerol fraction, the odd-carbon [U-(14)C]oleate also formed from [(14)C] acetate was in the polar lipid fraction. Kinetic data showed that this oleate was not channeled into cis-11-eicosenoate nor cis-13-docosenoate over a 24-hour period. Under suitable conditions, nasturtium seed could also produce [(14)C]stearate, [(14)C]eicosenoate, and [(14)C]docosenoate from [1-(14)C]acetate. The results are discussed in terms of the number of pathways producing fatty acids. From pool size and other considerations, the results can be rationalized only in terms of different de novo systems for oleate biosythesis, one supplying oleate for incorporation into phospholipids and the other supplying oleate for chain elongation and subsequent esterification into triacylglycerols. Because of the probable heterogeneous nature of the seed tissue, it is not known if these two systems are operating in different cell types, in the same cell type at different stages of development, or in the same cell type concurrently.

Biosynthesis of Acyl Lipids Containing Very-Long Chain Fatty Acids in Microspore-Derived and Zygotic Embryos of Brassica

Biosynthesis of very long chain (>C18) fatty acids (VLCFAs) and the pathway for their incorporation into acyl liipids was studied in microspore-derived (MD) and zygotic embryos of Brassica napus L. cv Reston. In the presence of [1-'4C]oleoyl-coenzyme A or [1-14C] eicosenoyl-coenzyme A, malonyl-coenzyme A, and reducing equivalents, maximal in vitro elongation activity was expressed in protein pr...

Chemical Survey and Erucic Acid Content of Commercial Varieties of Nasturtium , Tropaeolum maju 5 L .

Nasturtium (Tropaeolum majus L.) oil contains the highest levels of erucic acid of known seed oils (75-80%). A significant portion of the acid is attached to the 2-position of the glycerol, and trierucin is a major component (ca. 50%) of the oil. Seeds from eleven varieties of commercially available garden nasturtium (T. majus) were screened for oil content, erucic acid levels and fatty acid di...

Seed-specific heterologous expression of a nasturtium FAE gene in Arabidopsis results in a dramatic increase in the proportion of erucic acid.

The fatty acid elongase [often designated FAE or beta-(or 3-) ketoacyl-CoA synthase] is a condensing enzyme and is the first component of the elongation complex involved in synthesis of erucic acid (22:1) in seeds of garden nasturtium (Tropaeolum majus). Using a degenerate primers approach, a cDNA of a putative embryo FAE was obtained showing high homology to known plant elongases. This cDNA co...

Biosynthesis of triacylglycerols containing very long chain monounsaturated acyl moieties in developing seeds.

Particulate (15,000g) fractions from developing seeds of honesty (Lunaria annua L.) and mustard (Sinapis alba L.) synthesize radioactive very long chain monounsaturated fatty acids (gadoleic, erucic, and nervonic) from [1-(14)C]oleoyl-CoA and malonyl-CoA or from oleoyl-CoA and [2-(14)C]malonyl-CoA. The very long chain monounsaturated fatty acids are rapidly channeled to triacylglycerois and oth...

Different dietary omega-3 sources during pregnancy and DHA in the developing rat brain

Dietary sources of the essential fatty acid a-linolenic acid (ALA; 18:3n-3) include green leaves, some seeds, nuts and cooking oils. The principal dietary source of the long-chain (LC) n-3 polyunsaturated fatty acids (PUFA) eicosapentaenoic acid (EPA; 20:5n-3), docosapentaenoic acid (DPA; 22:5n-3) and docosahexaenoic acid (DHA; 22:6n-3) is oily fish, yet it is estimated that only 27% of UKadult...
