چکیده

The results of experiments designed to test the hypothesis that near-surface molecular structure of iron oxide minerals influences adhesion of dissimilatory iron reducing bacteria are presented. These experiments involved the measurement, using atomic force microscopy, of interaction forces generated between Shewanella oneidensis MR-1 cells and single crystal growth faces of iron oxide minerals. Significantly different adhesive force was measured between cells and the 001 face of hematite, and the 100 and 111 faces of magnetite. A role for electrostatic interactions is apparent. The trend in relative forces of adhesion generated at the mineral surfaces is in agreement with predicted ferric site densities published previously. These results suggest that near-surface structure does indeed influence initial cell attachment to iron oxide surfaces; whether this is mediated via specific cell surface-mineral surface interactions or by more general interfacial phenomena remains untested. © 2005 American Institute of Physics. Bioreduction of hematite nanoparticles by the dissimilatory iron reducing bacterium Shewanella oneidensis MR-1

We examined the reduction of different size hematite (a-Fe2O3) nanoparticles (average diameter of 11, 12, 30, 43, and 99 nm) by the dissimilatory iron reducing bacteria (DIRB), Shewanella oneidensis MR-1, to determine how S. oneidensis MR-1 may utilize these environmentally relevant solid-phase electron acceptors. The surface-area-normalized-bacterial Fe(III) reduction rate for the larger nanop...

Kinetic characterization of OmcA and MtrC, terminal reductases involved in respiratory electron transfer for dissimilatory iron reduction in Shewanella oneidensis MR-1.

We have used scaling kinetics and the concept of kinetic competence to elucidate the role of hemeproteins OmcA and MtrC in iron reduction by Shewanella oneidensis MR-1. Second-order rate constants for OmcA and MtrC were determined by single-turnover experiments. For soluble iron species, a stopped-flow apparatus was used, and for the less reactive iron oxide goethite, a conventional spectrophot...

Specific bonds between an iron oxide surface and outer membrane cytochromes MtrC and OmcA from Shewanella oneidensis MR-1.

Shewanella oneidensis MR-1 is purported to express outer membrane cytochromes (e.g., MtrC and OmcA) that transfer electrons directly to Fe(III) in a mineral during anaerobic respiration. A prerequisite for this type of reaction would be the formation of a stable bond between a cytochrome and an iron oxide surface. Atomic force microscopy (AFM) was used to detect whether a specific bond forms be...

Molecular Underpinnings of Fe(III) Oxide Reduction by Shewanella Oneidensis MR-1

In the absence of O(2) and other electron acceptors, the Gram-negative bacterium Shewanella oneidensis MR-1 can use ferric [Fe(III)] (oxy)(hydr)oxide minerals as the terminal electron acceptors for anaerobic respiration. At circumneutral pH and in the absence of strong complexing ligands, Fe(III) oxides are relatively insoluble and thus are external to the bacterial cells. S. oneidensis MR-1 an...

Directed Natural Forces of Affinity between a Bacterium and Mineral

The formation of a bond between a bacterium and a mineral is ultimately controlled by forces that operate over length scales of a few nanometers. This manuscript presents evidence that bacteria may actively modulate forces at the cell-mineral interface to promote contact with specific mineral phases. Nanoto pico-Newton forces were measured between goethite (FeOOH) and each of two species of Gra...
