چکیده

Galectin-9 expression in endothelial cells can be induced in response to inflammation. However, the mechanism of its expression remains unclear. In this study, we found that interferon gamma (IFN-γ) induced galectin-9 expression in human endothelial cells in a time-dependent manner, which coincided with the activation of histone deacetylase (HDAC). When endothelial cells were treated with the HDAC3 inhibitor, apicidin, or shRNA-HDAC3 knockdown, IFN-γ-induced galectin-9 expression was abolished. Over-expression of HDAC3 induced the interaction between phosphoinositol-3 kinase (PI3K) and interferon response factor 3 (IRF3), leading to IRF3 phosphorylation, nuclear translocation and galectin-9 expression. HDAC3 functioned as a scaffold protein for PI3K/IRF3 interaction. In addition to galectin-9 expression, IFN-γ also induced galectin-9 location onto plasma membrane, which was HDAC3 independent. Importantly, HDAC3 was essential for the constitutive transcription of PI3K and IRF3, which might be responsible for the basal level of galectin-9 expression. The phosphorylation of IRF3 was essential for galectin-9 expression. This study provides new evidence that HDAC3 regulates galectin-9 expression in endothelial cells via interaction with PI3K-IRF3 signal pathway.

HDAC Inhibition in Vascular Endothelial Cells Regulates the Expression of ncRNAs

While clinical and pre-clinical trials indicate efficacy of histone deacetylase (HDAC) inhibitors in disease mediated by dynamic lysine modification, the impact on the expression of non-coding RNAs (ncRNAs) remains poorly understood. In this study, we investigate high throughput RNA sequencing data derived from primary human endothelial cells stimulated with HDAC inhibitors suberanilohydroxamic...

P-117: Gene Expression and Developmental State of Mouse Cloned Embryos after Treatment with Histone Deacetylase Inhibitor,Suberoylanilide Hydroxamic Acid (SAHA)

Background: It is known that acetylation level of the nuclear histones in cloned embryos is lower compare to normally developed embryos. Histone deacetylas inhibitors (HDACi) with improvement of acetylation level in these embryos can affect embryo quality in pre-implantation stage and expression level of different genes especially developmental genes. Materials and Methods: In this research, SA...

Statins control oxidized LDL-mediated histone modifications and gene expression in cultured human endothelial cells.

OBJECTIVE Activation of the endothelium by oxidized low-density lipoprotein (oxLDL) has been implicated in the development of atherosclerosis. Histone modifications impact on the transcriptional activity state of genes. We tested the hypothesis that oxLDL-induced inflammatory gene expression is regulated by histone modifications and experienced the effect of statins on these alterations. METH...

Effects of Valproic Acid, a Histone Deacetylase Inhibitor, on improvement of Locomotor Function in Rat Spinal Cord Injury Based on Epigenetic Science

Background: The primary phase of traumatic spinal cord injury (SCI) starts by a complex local inflammatory reaction such as secretion of pro-inflammatory cytokines from microglia and injured cells that substantially contribute to exacerbating pathogenic events in secondary phase. Valproic acid (VPA) is a histone deacetylase inhibitor. Acetylation of histones is critical to cellular inflammatory...
