چکیده

1. Propylene oxide reacts with DNA in aqueous buffer solution at about neutral pH to yield two principal products, identified as 7-(2-hydroxypropyl)guanine and 3-(2-hydroxypropyl)adenine, which hydrolyse out of the alkylated DNA at neutral pH values at 37 degrees C. 2. These products were obtained in quantity by reactions between propylene oxide and guanosine or adenine respectively. 3. The reactions between propylene oxide and adenine in acetic acid were parallel to those between dimethyl sulphate and adenine in neutral aqueous solution; the alkylated positions in adenine in order of decreasing reactivity were N-3, N-1 and N-9. A method for separating these alkyladenines is described. 4. Deoxyguanylic acid sodium salt was alkylated at N-7 by propylene oxide in neutral aqueous solution. 5. The nature of the side chain in the principal alkylation products was established by mass spectrometry, and the nature of the products is consistent with their formation by the bimolecular reaction mechanism.

The reaction of mustard gas with nucleic acids in vitro and in vivo.

Mustard gas [di-(2-chloroethyl) sulphide] and analogous alkylating agents produce a wide range of biological effects (cf. review by Boyland, 1948). Some ofthese effects, such as induction ofmutations (Auerbach & Robson, 1946) and interference with mitosis and chromosome breakage (Darlington & Koller, 1947), stimulated interest in the reactions of alkylating agents with deoxyribonucleic acid. Pr...

Preparation and properties of some nicotinamide adenine dinucleotide analogues with pentose and purine modifications.

The structure of nicotinamide adenine dinucleotide was proved conclusively by chemical synthesis (1) and isolation (2) of the biologically active coenzyme from a reaction in which the 5,5’dinucleoside pyrophosphate was formed by condensing together the two component 5’-mononucleotides, adenosine monophosphate and nicotinamide mononucleotide, with dicyclohexylcarbodiimide or trifluoroacetic acid...

Thermal Properties of Deoxyribonucleic Acids from Sensitive and Resistant Plasmacytomas1

cate that cellular deoxyribonucleic acid (DNA) might be the primary and perhaps the critical site of alkylation by cytotoxic alkylating agents (21). The biologic systems used in most of these investigations, however, have not permitted a direct correlation between alkylation of DNA and the observed biologic effects; that is, there is a lack of evidence whether alkylation of DNA is essential to ...
