چکیده

A method is described in detail for the preparation of monospecific sera for typing brucella cultures. The importance of using entirely smooth cultures throughout the procedure is emphasized. A single intravenous injection of living brucellae in rabbits caused an active infection and the early appearance of high-titre antibodies. Sera obtained five to seven days after inoculation contained a higher proportion of specific antibodies than serum obtained ten days after inoculation. Trial absorptions of each serum sample were performed to determine the relative content of homologous and heterologous antibodies, so that the optimum proportions of absorbing cells to serum could be employed for the final absorption. In this manner sera were obtained which at a 1/160 dilution agglutinated the homologous antigen and at a 1/10 dilution did not agglutinate the heterologous antigen.Monospecific sera were used in typing 180 brucella strains isolated in Great Britain and other countries, and over 3000 cultures recovered from experimental infections. With the exception of two strains of Br. melitensis, all cultures were agglutinated to titre with only one of the monospecific sera.

Immunogens of Brucella Abortus S19 Identified By Two-Dimensional Gel Electrophoresis and Immunoblotting

Background: Lipopolysaccharides (LPSs) and several antigenic proteins of Brucella have been considered for preparation of diagnostic reagents and subunit vaccines. The objective of this study was to identify and compare immunogens of B. abortus S19 which induce humoral immune responses in human, goat and rabbit. Material and Methods: The bacterial whole cell extract was prepared in extraction b...

High Level Expression of Recombinant Ribosomal Protein (L7/L12) from Brucella abortus and Its Reaction with Infected Human Sera

Brucellosis, caused by Brucella spp., is an important zoonotic disease that causes abortion and infertility in cattle and undulant fever in humans. Various studies have examined cell-free native and recombinant proteins as candidate protective antigens in animal models. Among Brucella immunogenes, antigen based on ribosomal preparation has been widely investigated. In this study, the immunogeni...

Surface antigens of smooth brucellae.

Surface antigens of smooth brucellae were extracted by ether-water, phenol-water, trichloroacetic acid, and saline and examined by immunoelectrophoresis and gel diffusion with antisera from infected and immunized rabbits. Ether-water extracts of Brucella melitensis contained a lipopolysaccharide protein component, which was specific for the surface of smooth brucellae and was correlated with th...

Rapid identification of rough Brucella isolates by a latex coagglutination assay with the 25-kilodalton outer membrane protein and rough-lipopolysaccharide-specific monoclonal antibodies.

A latex coagglutination assay was developed to identify rough (R) isolates of Brucella. Latex beads were coated, via protein A, with either an anti-Brucella rough-lipopolysaccharide (R-LPS) monoclonal antibody (MAb) or an anti-Brucella 25-kDa outer membrane protein (Omp25) MAb. Slide agglutination tests were done for 68 strains of Brucella spp., including type strains of all biovars as well as ...
