چکیده

In central neurons, action potentials (APs) originate in the axon initial segment (AIS) presumably due to its high concentration of voltage-dependent sodium (NaV) channels. In the AIS, NaV and their cytoskeletal anchoring proteins display a highly periodical nanoscale arrangement with unknown functional relevance. In mouse hippocampal neurons, we examined a mutation of the AIS-specific protein IV-spectrin and assessed AP initiation locus, AIS molecular organization, and the precision of in vivo-like AP encoding. In mutants, NaV concentration was strongly reduced in AIS but not soma, while AIS nanoscale organization was preserved. Surprisingly, APs still originated in the axon, even if the axonal channel density was as low as in the somatic membrane. However, the timing of those APs was less precise, therefore the neuron’s encoding bandwidth was substantially decreased. Therefore, the high NaV density in the AIS is not required for axonal AP initiation, it is crucial for AP timing precision.

Action potential initiation and propagation in CA3 pyramidal axons.

Thin, unmyelinated axons densely populate the mammalian hippocampus and cortex. However, the location and dynamics of spike initiation in thin axons remain unclear. We investigated basic properties of spike initiation and propagation in CA3 neurons of juvenile rat hippocampus. Sodium channel alpha subunit distribution and local applications of tetrodotoxin demonstrate that the site of first thr...

Relative contributions of axonal and somatic Na channels to action potential initiation in cerebellar Purkinje neurons.

Neuronal excitability is likely to be regulated by the site of action potential initiation, the location on a neuron that crosses threshold first. Although initiation is axonal in many neurons, in Purkinje cells, somatic conductances can generate spontaneous action potentials, suggesting that the perisomatic region (soma and/or initial segment) contributes to spike initiation and may regulate f...

The right delay Detecting spike patterns with STDP and axonal delays

Axonal conduction delays are often ignored in simulations of spiking neural networks. Here, models of spiking neural networks with spiketiming-dependent plasticity, and conduction delays, are used in three simulation experiments. The first two are based on studies by Masquelier et al. [10, 11], and the third is an expansion on their work. First, it is confirmed that a neuron can learn to detect...

Polarized distribution of ion channels within microdomains of the axon initial segment.

Voltage-gated sodium (Na(v)) channels accumulate at the axon initial segment (IS), where their high density supports spike initiation. Maintenance of this high density of Na(v) channels involves a macromolecular complex that includes the cytoskeletal linker protein ankyrin-G, the only protein known to bind Na(v) channels and localize them at the IS. We found previously that Na(v)1.6 is the pred...

Action potential fidelity during normal and epileptiform activity in paired soma-axon recordings from rat hippocampus.

Although action potential initiation and propagation are fundamental to nervous system function, there are few direct electrophysiological observations of propagating action potentials in small unmyelinated fibres, such as the axons within mammalian hippocampus. To circumvent limitations of previous studies that relied on extracellular stimulation, we performed dual recordings: whole-cell recor...
