Response: Cell fate in the early mouse embryo--sorting out the influence of developmental history on lineage choice.

چکیده
The first three cell lineages formed in the implanting mouse embryo are the epiblast (EPI), primitive endoderm (PE) and trophectoderm. The EPI and PE are the two lineages derived from the inner cell mass (ICM) of the blastocyst, which give rise to the fetus and extraembryonic yolk sac, respectively. Previous studies suggest that embryonic day 3.5 ICM cells begin as a mixture of PE or EPI progenitors randomly distributed in the ICM which then sort out during blastocyst expansion to form two morphologically distinct layers. How are these two progenitors first specified? A proposed model is that the divisional history of ICM cells affects EPI/PE lineage specification. Two independent research groups addressed this issue using completely different approaches and reached different conclusions (Morris et al., 2010; Yamanaka et al., 2010). What causes the apparent discrepancy between the two studies? This commentary highlights two important differences in the design of the two studies: the timing of EPI/PE lineage evaluation and the strategy for scoring the results. It is proposed that these studies are not in conflict, but rather are complementary.

Formation of distinct cell types in the mouse blastocyst.
Early development of the mouse comprises a sequence of cell fate decisions in which cells are guided along a pathway of restricted potential and increasing specialisation. The first choice faced by cells of the embryo is whether to become trophectoderm (TE) or inner cell mass (ICM); TE is an extra-embryonic tissue which will form the embryonic portion of the placenta, whilst ICM gives rise to c...
متن کاملP-26: The Effect of Zygote and 2-cell Development Stages on Vitrification Process of Mouse Embryo
Background: While it is possible to routinely cryopreserve embryos from several mammalian species, the cryopreservation of embryos has largely been limited by their high sensitivity to chilling injury. Many factors such as the stage of embryonic development, cryoprotectant toxicity, the composition of the vitrification solution and cooling and warming rates can influence survival of embryos aft...
متن کاملTime Course of Degradation and Deadenylation of Maternal c-mos and Cyclin A2 mRNA during Early Development of One-Cell Embryo in Mouse
Early in the development of many animals, before transcription begins, any change in the pattern of protein synthesis is attributed to a change in the translational activity or stability of mRNA in the egg and early embryo. As a result, translational control is critical for a variety of developmental decisions, including oocyte maturation and initiation of preimplantation development. In this s...
متن کاملP-115: Melatonin Increases Developmental Rate of In Vitro Mouse Somatic Cell Nuclear
Background: The beneficial effect of supplementing culture medium with melatonin has been reported during in vitro embryo development of species such as mouse, bovine and porcine. However, the effect of melatonin on the mouse somatic cell nuclear transfer remained unknown. Materials and Methods: In this study, we assessed the effects of various concentrations of melatonin (10-6 to 10-12 M) on t...
متن کاملDetermination the Effects of Epidermal Growth Factor on Developmental Rates of Pre-Implantation Mouse Embryo
Purpose: To determine the effects of different doses of addition of exogenous epidermal growth factor (EGF) on development of pre-implantation mouse embryo. Materials and Methods: Mouse zygotes, two cell, 8 cell any morulae were collected from superovulated NMRI mice 24, 48, 64 and 80 hrs after hCG injection, respectively. The obtained embryos were cultured on medium alone, medium with 1, 4 an...
