چکیده

This study examines mechanisms that regulate T helper lymphocyte (Th) 1 commitment in Helicobacter pylori-infected human gastric mucosa. Interferon (IFN)-γ, interleukin (IL)-4, and IL-12 were measured in total extracts of gastric biopsies taken from H. pylori-infected and uninfected patients by enzyme linked immunosorbent assay. STAT4, STAT6, and Tbet were examined in total proteins extracted from gastric biopsies by Western blotting. Finally, the effect of a neutralizing IL-12 antibody on the expression of Th1 transcription factors and levels of IFN-γ was examined in organ cultures of H. pylori-infected biopsies. Increased levels of IFN-γ and IL-12 were found in gastric biopsy samples of H. pyloriinfected patients compared to uninfected patients. Accordingly, H. pylori-infected biopsies exhibited high expression of phosphorylated STAT4 and T-bet. Higher levels of IFN-γ and expression of Th1 transcription factors were associated with higher infiltration of mononuclear cells in the gastric mucosa. By contrast, production of IL-4 and expression of phosphorylated STAT6 were not associated with the intensity of mononuclear cell infiltration. In ex vivo organ cultures of H. pylori-infected biopsies, neutralization of endogenous IL-12 down-regulated the expression of p-Stat4 and T-bet and reduced IFN-γ production. Data indicate that IL-12 contributes to the Th1 cell commitment in H. pylori-infected human gastric mucosa. AC CE PT ED on A uust 9, 2017 by gest http/iai.asm .rg/ D ow nladed fom

Interleukin-12 drives the Th1 signaling pathway in Helicobacter pylori-infected human gastric mucosa.

In this study we examined mechanisms that regulate T-helper lymphocyte 1 (Th1) commitment in Helicobacter pylori-infected human gastric mucosa. The levels of gamma interferon (IFN-gamma), interleukin-4 (IL-4), and IL-12 in total extracts of gastric biopsies taken from H. pylori-infected and uninfected patients were determined by an enzyme-linked immunosorbent assay. The levels of signal transdu...

Expression of Interleukin-18, a Th1 cytokine, in human gastric mucosa is increased in Helicobacter pylori infection.

Interleukin-18 (IL-18), a cytokine that promotes Th1 responses, is processed to the active mature protein by caspase-1. The effects of Helicobacter pylori infection on gastric IL-18 and caspase-1 were examined. In antral mucosa, IL-18 mRNA expression was greater (P<.01) in H. pylori-positive (n=40) than in H. pylori-negative patients (n=29) with normal mucosa. Inactive precursor (24 kDa) and ma...

Differential stimulation of interleukin-12 (IL-12) and IL-10 by live and killed Helicobacter pylori in vitro and association of IL-12 production with gamma interferon-producing T cells in the human gastric mucosa.

The objective of these experiments was to examine the ability of Helicobacter pylori to stimulate interleukin-10 (IL-10) or IL-12 and select for either Th1 or Th2 cells. Gastric biopsy specimens were collected from patients who were categorized with respect to the presence of H. pylori and gastric disease as well as their age, gender, medications, and other factors. As Th1 and Th2 cells are sel...

Enhanced disease severity in Helicobacter pylori-infected mice deficient in Fas signaling.

Recent evidence suggests that immune-mediated gastric epithelial cell apoptosis through Fas-Fas ligand interactions participates in Helicobacter pylori disease pathogenesis. To define the role of Fas signaling in vivo, H. pylori strain SS1 infection in C57BL/6 mice was compared to that in mice deficient in the Fas ligand (gld). gld mice had a degree of gastritis similar to that of C57BL/6 mice ...

Association between Helicobacter pylori cagA, babA2 Virulence Factors and Gastric Mucosal Interleukin-33 mRNA Expression and Clinical Outcomes in Dyspeptic Patients

Helicobacter pylori (H. pylori) infection has been reported in more than half of the world human population. It is associated with gastric inflammation and noticeable infiltration of the immune cells to the stomach mucosa by several cytokines secretion. IL-1&beta, IL-18 have been shown to contribute to H. pylori induced gastritis, but the details of inflammation and association of virulence fac...
