چکیده

Three connexin genes have been reported to be expressed in the lens. The a1 connexin (Cx43) has been detected in mouse lens epithelial cells (Yancey et al., Xiaohua Gong,* En Li,† George Klier,* Qingling Huang,‡ Ying Wu,* Hong Lei,† Nalin M. Kumar,* Joseph Horwitz,‡ and Norton B. Gilula*§ 1992). Both a3 (Cx46) and a8 (Cx50) connexin have been detected in the elongating primary fibers, as well as in *Department of Cell Biology The Scripps Research Institute the secondary fibers during lens development, and they are colocalized to the same gap junction plaques (Paul La Jolla, California 92037 †Cardiovascular Research Center, MGH et al., 1991). The most abundant proteins in the vertebrate lens are Harvard Medical School Charlestown, Massachusetts 02129 crystallins, which account for more than 90% of the lens-soluble proteins (Bloemendal, 1981; Harding and ‡Jules Stein Eye Institute at UCLA Los Angeles, California 90095 Crabbe, 1984). Crystallins from mammalian lenses are divided into three major classes: a-, b-, and g-crystallins. Lens transparency results from the suprastructural organization of the lens that is created by both plasma Summary membrane components and cytoplasmic proteins, including the crystallins. However, the molecular mechaGap junction channels formed by a3 (Cx46) and a8 nisms by which this organization is generated and main(Cx50) connexin provide pathways for communication tained are not well understood. between the fiber cells in the normal transparent lens. SPARC deficiency leads to early-onset cataractogenesis.

PURPOSE To determine the role of SPARC (secreted protein, acidic, and rich in cysteine) in cataractogenesis by examining mice deficient in a matricellular protein SPARC. METHODS Mice were rendered SPARC-deficient by a targeted disruption of the gene. Slit-lamp microscopy and histology were used to examine the eyes of SPARC-null and wild-type mice from birth to 14 months of age. RESULTS SPAR...

Contribution of calpain Lp82-induced proteolysis to experimental cataractogenesis in mice.

PURPOSE The purpose of the present experiments was to provide a biochemical mechanism for the involvement of lens-specific calpain Lp82 in experimental cataractogenesis in mice. METHODS Nuclear cataracts were produced by culturing lenses from 4-week-old mice and rats in calcium ionophore A23187 or by injection of buthionine sulfoximine (BSO) into 7-day-old mice. Casein zymography, sodium dode...

Gene disruption in Salmonella typhimurim by modified λ Red disruption system

There are many techniques to knock out directed genes in bacteria, some of which have been described in Salmonella species. In this study, a combination of SOEing PCR method and the λ Red disruption system were used to disrupt phoP gene in wild type and standard strains of Salmonella typhimurium. Three standards PCR and one fusion PCR reactions were performed to construct a linear DNA including...

Defining a link between gap junction communication, proteolysis, and cataract formation.

Disruption of the connexin alpha 3 (Cx46) gene (alpha 3 (-/-)) in mice results in severe cataracts within the nuclear portion of the lens. These cataracts are associated with proteolytic processing of the abundant lens protein gamma-crystallin, leading to its aggregation and subsequent opacification of the lens. The general cysteine protease inhibitor, E-64, blocked cataract formation and gamma...
