Thioredoxin Reductase is More Effective than Glutathione Reductase in Inducing Protection against Plasmodium Berghei Infection
نویسنده
چکیده
Thioredoxin reductase and glutathione reductase are important enzymes of redox system that help the malaria parasite to maintain an adequate intracellular redox environment and contribute greatly to the antioxidant capacity of the cell. In the present study humoral immune response directed against thioredoxin reductase and glutathione reductase during Plasmodium berghei infection were investigated. Thioredoxin reductase and glutathione reductase induced humoral immune response in Balb/c mice immunized with purified parasite protein using saponin. Pre-challenge sera gave antibody titre of 1:512. IFA showed specificity of antimalarials antibodies towards parasite. Immunization with thioredoxin reductase provides partial protection compared to glutathione reductase in mice to P. berghei infection. Immunization with thioredoxin reductase delays the progression of infection as compared to

Glutathione reductase and thioredoxin reductase: novel antioxidant enzymes from Plasmodium berghei.
Malaria parasites adapt to the oxidative stress during their erythrocytic stages with the help of vital thioredoxin redox system and glutathione redox system. Glutathione reductase and thioredoxin reductase are important enzymes of these redox systems that help parasites to maintain an adequate intracellular redox environment. In the present study, activities of glutathione reductase and thiore...

Thioredoxins (Trxs) are small ubiquitous oxidoreductase proteins with two redox-active Cys residues in a conserved active site (WCG/PPC) that regulate numerous target proteins via thiol/disulfide exchanges in the cells of prokaryotes and eukaryotes. The isoforms OsTrx23 with a typical active site (WCGPC) and OsTrx20 with an atypical active site (WCTPC) are two Trx h- type isoforms in rice that ...
متن کاملCompounds structurally related to ellagic acid show improved antiplasmodial activity.
The cancer chemopreventive agent ellagic acid (EA) is a known inhibitor of glutathione S-transferases (GSTs) and possesses antiplasmodial activities in the upper-nanomolar range. In the recent drug development approach, the properties of the active site of Plasmodium falciparum GST were exploited for inhibitor design by introducing one or two additional hydroxyl groups into EA, yielding flavell...
متن کاملMethylseleninate is a substrate rather than an inhibitor of mammalian thioredoxin reductase. Implications for the antitumor effects of selenium.
Biochemical and clinical evidence indicates that monomethylated selenium compounds are crucial for the tumor preventive effects of the trace element selenium and that methylselenol (CH(3)SeH) is a key metabolite. As suggested by Ganther (Ganther, H. E. (1999) Carcinogenesis 20, 1657-1666), methylselenol and its precursor methylseleninate might exert their effects by inhibition of the selenoenzy...
متن کاملGlutathione and thioredoxin systems of the malaria parasite Plasmodium falciparum: Partners in crime?
In P. falciparum, antioxidant proteins of the glutathione and thioredoxin systems are compartmentalized. Some subcellular compartments have only a partial complement of these proteins. This lack of key anti-oxidant proteins in certain sub-cellular compartments might be compensated by functional complementation between these systems. By assessing the cross-talk between these systems, we show for...
