New chromogenic substrate for determination of serum amylase activity.

چکیده
A rapid assay for serum amylase activity has been developed based on the use of a new chromogenic substrate-Cibachron Blue-amylose. The procedure requires 0.1 ml of serum and measures the production of soluble chro. mogen, the formation of which islinearwith enzyme activity. Normal human serum has a mean amylase activity of 118 ± 37 (SD) mg dye/100 ml/15 mm or, in international units, 78 ± 24 (SD) g dye/mm/mI. Excellent correlation was observed with a saccharometric amylase procedure.

Comparison of two methods for determining amylase activity in serum and urine.
E CLASSIC measurement of a-amylase (a1,4-glucan 4-glucanohydrolase, EC 3.2.1.1) activity in biological fluids is based on enzymatic hydrolysis of a starch substrate. This activity can be assessed either by a saccharogenic procedure (1, ), which measures the increase in reducing sugar (mainly maltose), or by an amyloclastic procedure (1, ), which depends on the decrease or disappearance of the b...
متن کاملBiological Activity Analysis of Native and Recombinant Streptokinase Using Clot Lysis and Chromogenic Substrate Assay
Determination of streptokinase activity is usually accomplished through two assay methods: a) Clot lysis, b) Chromogenic substrate assay. In this study the biological activity of two streptokinase products, namely Streptase®, which is a native product and Heberkinasa®, which is a recombinant product, was determined against the third international reference standard using the two forementioned a...

متن کاملKinetic nephelometric procedure for measurement of amylase activity in serum.
We developed a kinetic procedure for determination of amylase activity in serum by use of nephelometric measurements. Light scattering from the substrate, a stable suspension of starch, is decreased as amylase hydrolyzes the starch to soluble fragments. Values obtained for serum amylase correlate closely with those determined by a method in which a starch-dye complex is used. Precision of the n...
متن کاملDirect SpectrophotometricDeterminationof a-Amylase Activityin Saliva, with p-Nitrophenyla-Maltoside as Substrate
We describe a simple, direct kinetic method for determination of salivary a-amylase (1,4 a-o-glucan 4-glucanohydrolase, EC 3.2.1.1). The assay makes use of a well-defined substrate, p-nitrophenyl a-maltoside, which is hydrolyzed by a-amylase to a chromogenic product, p-nitrophenol. Activity is determined by directly monitoring the increase in absorbance of the reaction mixture. Amylase activity...
