چکیده

Upon entry into the central nervous system (CNS), serum insulin-like growth factor-1 (IGF-I) modulates neuronal growth, survival and excitability. Yet mechanisms that trigger IGF-I entry across the blood-brain barrier remain unclear. We show that neuronal activity elicited by electrical, sensory or behavioral stimulation increases IGF-I input in activated regions. Entrance of serum IGF-I is triggered by diffusible messengers (i.e. ATP, arachidonic acid derivatives) released during neuro-vascular coupling. These messengers stimulate matrix metalloproteinase-9, leading to cleavage of the IGF binding protein-3 *Manuscript Click here to view linked References 2 (IGFBP-3). Cleavage of IGFBP-3 allows the passage of serum IGF-I into the CNS through an interaction with the endothelial transporter lipoprotein related receptor 1. Activity-dependent entrance of serum IGF-I into the CNS may help to explain disparate observations such as pro-neurogenic effects of epilepsy, rehabilitatory effects of neural stimulation and modulatory effects of blood flow on brain activity.

Brain-derived neurotrophic factor, insulin-like growth factor-I and its binding protein responses to a session of endurance exercise in healthy elderly men

Purpose: This study investigated the effect of endurance activity on brain-derived neurotrophic   factor(BDNF), insulin-like growth factor 1(IGF-1) and its binding protein 3(IGFBP-3) in elderly   healthy individuals.   Materials and Methods: Eleven healthy old males (mean age of 68 ± 2.31 years old, height   of 177 ± 3.1 cm and weight of 79 ± 1.5 kg) were studied. Seventy two hours after maxi...

P-64: The Relationship between Polymorphism in Gene of Insulin-Like Growth Factor-I and The Serum Periparturient Concentration in Holstein Dairy Cows

Background: One of the most important metabolic factors affecting the reproductive activity is insulin-like growth factor-I (IGFI) concentration changes after calving. Recently, Maj et al. (2008) discovered a significant association between the IGF-I genotypes based on the 5'-untranslatedregion (5'-UTR) of IGF-I gene and the IGF-I blood level. The objective of this study is to investi...

Insulin and insulin-like growth factor receptors in the brain: Physiological and pathological aspects

The involvement of insulin, the insulin-like growth factors (IGF1, IGF2) and their receptors in central nervous system development and function has been the focus of scientific interest for more than 30 years. The insulin-like peptides, both locally-produced proteins as well as those transported from the circulation into the brain via the blood-brain barrier, are involved in a myriad of biologi...

The Brain Response to Peripheral Insulin Declines with Age: A Contribution of the Blood-Brain Barrier?

OBJECTIVES It is a matter of debate whether impaired insulin action originates from a defect at the neural level or impaired transport of the hormone into the brain. In this study, we aimed to investigate the effect of aging on insulin concentrations in the periphery and the central nervous system as well as its impact on insulin-dependent brain activity. METHODS Insulin, glucose and albumin ...
