چکیده
Ascorbate Determination, 2,4-Dinitrophenylhydrazine Microamounts of vitamin C could be readily determined in 20 ul-samples using the 2,4-dinitrophenylhydrazine method together with separation by thin layer chromato graphy. The condensation reaction was carried out for 5 min at 100 °C on a glass fibre disc. Purification of vitamin C hydrazones was accomplished by repeated separation on TLC plates. An aqueous solution of 65% acetic acid was em ployed to dissolve the vitamin C hydrazones, providing m axi mal absorbance at 500 nm. The minimum amount detectable by this method is 0.4 i/g of dehydroascorbic acid. The macrodetermination of vitamin C was improved by sim pli fying a previous method and employing 65% aqueous acetic acid as a solvent for the hydrazones.

Thin-layer chromatographic determination of L-ascorbic, L-dehydroascorbic nd 2.3-diketo-L-gulonic acids in animal tissues, blood and urine.
A new method for the determination of ascorbic, dehydroascorbic and 2.3-diketogulonic acids in animal tissues, blood and urine is described. The material for analysis is homogenized in the presence of stannous chloride in trichloracetic acid and the extract is divided into three aliquots: in one part the ascorbic acid is converted into dehydroascorbic acid by oxidation with bromine water, in th...
متن کاملNew method for the estimation of platelet ascorbic acid.
Present techniques for the estimation of platelet ascorbic acid allow interference by other substances in the sample. A new and more specific method of analysis is presented. The proposed method owes its increased specificity to resolution of the extract by thin-layer chromatography. By this means ascorbic acid is separated from other reducing substances present. The separated ascorbic acid is ...
متن کاملComparative analyses for ascorbic acid by the 2,4-dinitrophenylhydrazine method with the coupling reaction at different temperatures: a procedure for determining specificity.
The dinitrophenylhydrazine method for the determination of ascorbic acid is based upon treatment with 85% HzS04 of the chromogen formed by the coupling of 2,4-dinitrophenylhydrazine with oxidized ascorbic acid. In the original publication of this method, Roe and Kuether (1) set the temperature of the coupling reaction at 37” and used a 3-hour incubation period. These authors showed that a low t...

0x0 compounds (2,4-dinitrophenylhydrazine-reactive) formed during the Maillard reaction between glucose and glycine were converted into 2,4_dinitrophenylhydrazones and were separated into five main fractions by reversed-phase chromatography on Separon SGX Cl8 using a methanol-water gradient. Four of these fractions were homogeneous, as shown by thin-layer chromatography. One of the fractions st...
متن کاملA rapid, simple method for the determination of reduced, dehydro-, and total ascorbic acid in biological material.
Chemical methods for the determination of reduced ascorbic acid (Zascorbic acid) are based upon its reducing properties. The indophenol method of Bessey (1, 2) and the perinaphthindantrione hydrate method of Moubasher (3) are examples of these methods. Modifications to increase the specificity of the indophenol method have been presented by Robinson and Stotz (4), Lowry, Lopez, and Bessey (5), ...
