Loblolly Pine Somatic Embryogenesis: Development of a Maturation Medium and Resulting Embryo Quality
نویسندگان
چکیده
Clonal production of loblolly pine (Pinus taeda L.) through somatic embryogenesis has great potential to meet the increasing industrial demands for highquality uniform raw materials. A major barrier to the commercialization of this technology is the low quality of resulting embryos. Twenty-five newly initiated loblolly pine genotypes were followed through the process of liquid culture establishment, embryo maturation, germination, and retrieval from cryogenic storage. A maturation medium, capable of promoting development of loblolly pine somatic embryos that can germinate, is presented that combines Y2 P6 modified salts, 2% maltose, 13% polyethylene glycol 8000, 5 mg/L abscisic acid, and 2.5 g/L Gelrite. A procedure for converting and acclimating germinants to growth in soil and greenhouse conditions is presented. A set of somatic seedlings, produced from the maturation medium, showed 100% survival when planted in a field setting. Somatic seedlings showed normal yearly growth when compared to standard seedlings from natural seed. Quality of resulting embryos is examined and compared to zygotic embryos using morphology, dry weight, germination performance, and gene expression. All observations support the conclusion that somatic embryos grow approximately halfway through the normal sequence of development and then prematurely discontinue growth.

An Embryo Staging System for Comparison of Zygotic and Somatic Embryo Development
An embryo classification system has been developed based on observations of Pseudotsuga menziesii (Mirb.) Franco (Douglas-fir) and Pinus taeda L. (loblolly pine) zygotic embryos and on the literature concerning embryology within the pine family. This system is suitable for classification of both zygotic and somatic embryos of loblolly pine, Picea abies L. (Norway spruce), and Douglas-fir. This ...

Background Brazilian pine (Araucaria angustifolia (Bert) O. Ktze) is the only native conifer species with economic importance in Brazil. Recently, due to intensive exploitation Brazilian pine was included in the official list of endangered Brazilian plants. Biotechnology tools, like somatic embryogenesis (SE), may become a potentially useful tool for mass clonal propagation and ex situ conserva...

Somatic embryogenesis is affected by several factors. In this research project, we studied the effect of explant size, wounding and desiccation treatments on somatic embryogenesis and their conversion into plantlet among three genotypes of soybean. The explants were sampled from immature embryos of soybean in three different sizes (3, 5 & 7 mm) with wounding treatment on half of each, and then ...

متن کاملSOMATIC EMBRYOGENESIS AND REGENERATION OF PLANTLET IN SAFFRON, CROCUS SATIVUS L.
Somatic embryogenesis was initiated in C. sativus L. from shoot meristems on LS medium containing BAP (2×10-5 M)+NAA (2×10-5 M). Various stages of somatic embryogenesis were observed in the same medium and the development was asynchronous. Somatic embryo development proceeded through well recognized sequences (globular to embryoids) with clearly discernible bipolar regions. Matured embryos coul...
