External quality control of urinary methyl malonic acid quantification - announcement of a pilot study.

چکیده
Methylmalonic acid (MMA) accumulates in the body in cases of cobalamin (vitamin B12) deficiency because of impaired metabolization of methylmalonyl CoA to succinyl CoA by methylmalonyl CoA-mutase, a cobalamin-dependent enzyme. Therefore, MMA represents a functional marker of cobalamin deficiency (1). It is widely accepted that MMA measurement is useful and necessary if serum cobalamin concentrations between 150 and 300 pg/mL are found, overcoming the limited reliability of serum cobalamin measurement in the lower concentration range (2–4). Despite good evidence for the usefulness of MMA measurement, tests are rarely requested in many countries and are offered by only a few laboratories. This particularly applies for measurement of urinary MMA, which clearly represents the sample material of choice; impaired renal function is associated with increased serum and plasma MMA concentrations, irrespective of the cobalamin status (5). In contrast, measurement of urinary MMA concentrations and calculation of the ratio of urinary MMA to urinary creatinine is essentially independent of individual renal function. Quantification of MMA in both serum and urine is technically demanding and gas chromatography (GC) or liquid chromatography (LC) isotope dilution mass spectrometry methods are mandatory. So far, no matrix-based materials for MMA assay calibration and continuous quality control are commercially available. After initial studies (6, 7), proficiency testing programs for serum and plasma MMA measurement were implemented in Europe and the USA. For urinary MMA measurement, however, no external quality control programs have been implemented so far.

External quality assessment of urinary methylmalonic acid quantification - results of a pilot study.
Measurement of methylmalonic acid (MMA) is recognised as a useful tool in the diagnostic work-up of suspected cobalamin deficiency (1). Increased concentrations of MMA in both serum and urine are indicative of cobalamin deficiency at the tissue level. MMA measurement in serum as the sample material is convenient from a logistic point of view, since analysis can be performed using residual sampl...
متن کاملUrine methyl malonic acid levels in HIV-infected adults with peripheral neuropathy
Background: Cobalamin deficiency and peripheral neuropathy (PN) are commonly seen in HIV-infected adults. The level of urine methylmalonic acid (UMMA), a reliable indicator of tissue cobalamin status, was determined in HIV infected subjects with and without PN to establish this association. Methods: One hundred and ninety-eight (198) consenting HIV infected subjects with and without PN were rec...
متن کاملTo ascertain the utility of urinary methyl malonic Acid as a potential marker of ischemic heart failure.
BACKGROUND Methyl Malonic Acid (MMA) is known to be an integral component of the cascade of events in mitochondrial energy metabolism and since heart failure involves energy pathways, it is probable that levels of MMA could be used as a reliable biomarker to objectively identify the disease during the early stages and help in prognostication. MATERIALS AND METHODS The present study was envisa...
متن کاملDevelopment of qRT-PCR Test for Quantification of Rubella Virus in Commercially Available Vaccines
vaccines is considered as a critical point and in-process of quality control (IPQC) test of vaccine production. Rapid tests, like real time PCR, are more appropriated when the production occurs at industrial scale because of the amounts of starting materials and the excess of consumed time required. In the current study, a real-time quantitative reverse transcription-polymerase chain reaction...
متن کاملRice Bran as a Sorbent for Malonic Acid From Aqueous Solution
The adsorption of malonic acid on the rice bran was studied under various conditions such as temperature, contact time, adsorbent dose and concentration of adsorbate. Batch adsorption experiments were conducted and the result showed that the adsorption was dependent on all these parameters. The adsorption process obeys the Langmuir and Freundlich adsorption isotherms. The adsorption capacity wa...
