چکیده

This paper describes a simple, kinetic model of the effect of an antibody on the binding of a toxin to its receptor. This model has been used to evaluate the contributions of antibody affinity and concentration to reduction in complex formation and enable prediction of the antibody kinetic constants and concentration required to provide a specified degree of protection. An expression for determining equilibrium time for the system has also been derived. Based on the proposed model we analytically calculated the relative reduction in toxin-receptor complex formation a variety of input parameters (kinetic constants and concentrations). Our analytical results were validated by numerical simulations, using the COPASI tool. Experimental validation of the model may provide a useful tool for in vitro selection of potentially therapeutic and prophylactic antibodies for progression to in vivo evaluation.

The receptor-toxin-antibody interaction: Mathematical model and numerical simulation

An antibody, also known as an immunoglobulin, is a protein used by the immune system to identify, neutralize, or kill foreign objects like bacteria, viruses, or pollen which are termed as antigen. The production of antibodies is the main function of the immune system. An antigen, when introduced into the body, triggers the production of an antibody by immune system which will then kill or neutr...

Alpha Toxin Purification and Antibody Production Against Local Strain of Clostridium septicum NH2

BACKGROUND: Clostridium septicum has played a significant role as a causative agent of many acute fetal diseases in man and animals. Alpha- toxin is the main factor in the pathogenesis of C. septicum with hemolytic, necrotic and lethal activities. OBJECTIVES: The study was designed to evaluate alpha-toxin purification and antibody production rate against a local strain of C. septicum NH2 which ...

Molecular and structural basis of the specificity of a neutralizing acetylcholine receptor-mimicking antibody, using combined mutational and molecular modeling analyses.

The antagonist activity of short-chain toxins from snake venoms toward the nicotinic acetylcholine receptor (nAChR) is neutralized upon binding to a toxin-specific monoclonal antibody called Malpha2-3 (1). To establish the molecular basis of this specificity, we predicted from both mutational analyses and docking procedures the structure of the Malpha2-3-toxin complex. From knowledge of the fun...

The Importance of α-CT and Salt bridges in the Formation of Insulin and its Receptor Complex by Computational Simulation

Insulin hormone is an important part of the endocrine system. It contains two polypeptide chains and plays a pivotal role in regulating carbohydrate metabolism. Insulin receptors (IR) located on cell surface interacts with insulin to control the intake of glucose. Although several studies have tried to clarify the interaction between insulin and its receptor, the mechanism of this interaction r...
