Synthesis of D-Glucose-1-C14 and D-Mannose-1-C14

چکیده
Heretofore, radiochemical yields of sugars position-labeled with carbon-14 have been only 5 to 10 percent. This paper reports an improved cyanohydrin synthesis, and the preparation of D-glucose-1-C and D-mannose-1-C from D-arabinose in yields of about 50 percent. Low yields in prior syntheses appear to have been due to several inefficient steps, and to the fact that extension of the carbon chain yields a pair of epimeric sugar derivatives, the proportion of which may not favor the desired epimer. In the classical cyanohydrin synthesis of glucose from D-arabinose only 30 percent of the product goes to the gluconic epimer. To increase the yield of glucose, a systematic study was made of the proportions of the epimers formed from D-arabinose under a variety of conditions. It was found that the ratio of the epimers depends upon the conditions under which the reaction of the sugar with cyanide takes place. Thus, D-arabinose and cyanide, in the presence of sodium bicarbonate and carbon dioxide, yield about three parts of mannonic nitrile to one part of gluconic nitrile, but, in the presence of sodium carbonate the proportion is approximately reversed.

Utilization of 1-C14-D-glucose by Torula utilis yeast.
A study of the utilization of l-C14-D-glucose by Torula utilis yeast was undertaken in an effort to clarify certain biosynthetic mechanisms operating in yeast metabolism. A recent publication by Gilvarg (1) describes the results obtained in a similar study with Xaccharomyces cerevisiae yeast. Since our methods and certain of our results differ from those of Gilvarg, a partial report of our obse...

In a previous publication (l), data were reported concerning n-mannose and n-ribose isolated from Torula utilis yeast which had been grown on n-glucose-l-C’4 as the sole source of carbon. It was found that water extraction of the lyophilized yeast yielded a mannan that was completely devoid of radioactivity. Further, the D-ribose obtained from the purine ribosides of the nucleic acid fraction s...

Although considerable research has been conducted in recent years on the mechanisms of fatty acid synthesis and oxidation in animal tissues (1) and in microorganisms (2), no similar studies of higher plants have yet been reported, despite the great importance of fat-storing seeds and fruits in the economy of nature and the value to comparative biochemistry of an understanding of fatty acid biog...
متن کاملSerum Levels of Acylcarnitines Are Altered in Prediabetic Conditions
OBJECTIVE The role of mitochondrial function in the complex pathogenesis of type 2 diabetes is not yet completely understood. Therefore, the aim of this study was to investigate serum concentrations of short-, medium- and long-chain acylcarnitines as markers of mitochondrial function in volunteers with normal, impaired or diabetic glucose control. METHODS Based on a 75 g oral glucose toleranc...

Lung surfactant mainly comprises phosphatidylcholines (PC), together with phosphatidylglycerols and surfactant proteins SP-A to SP-D. Dipalmitoyl-PC (PC16:0/16:0), palmitoylmyristoyl-PC (PC16:0/14:0), and palmitoylpalmitoleoyl-PC (PC16:0/16:1) together comprise 75-80% of surfactant PC. During alveolarization, which occurs postnatally in the rat, PC16:0/14:0 reversibly increases at the expense o...
