Chemical and physical properties of freezing point-depressing glycoproteins from Antarctic fishes.

چکیده
Glycoproteins were isolated from the blood sera of the Antarctic fishes Tremafomus borchgrevinki and Dissostichus mawsoni. Some of these glycoproteins have the unique property of depressing the freezing point of water more than expected on the basis of the number of particles present in solution and are termed active glycoproteins. These freezing point-depressing glycoproteins depress the freezing point of water to the same extent as NaCl on a weight basis. They are composed primarily of threonine (16%), alanine (23 %), N-acetylgalactosamine (29%), and galactose (28%). Compared with the active glycoproteins, the inactive ones (which form ideal solutions) have approximately the same composition except that proline is present. The active glycoproteins have been separated into three distinct groups, based on results from ion exchange chromatography and acrylamide gel electrophoresis. The three active glycoproteins occur in equal amounts in the blood sera, have identical compositions on a weight basis, but differ in molecularweights (10,500,17,000 and 21,500 g). Studies of viscosity and circular dichroism and dialysis experiments indicate that the active glycoproteins are expanded molecules, a property which undoubtedly is of importance to their function.

Protein and glycoprotein antifreezes in the intestinal fluid of polar fishes.
Measurements of ion concentrations, freezing points and melting points of intestinal fluid were made for several Antarctic fishes and two North Atlantic species. These measurements indicated that plasma and intestinal fluid are nearly isosmotic. Freezing points of intestinal fluid were approximately 0.9 degrees C below the melting points, suggesting the presence of glyco-protein antifreeze with...
متن کاملAntifreeze glycoprotein from an Antarctic fish. Effects of chemical modifications of carbohydrate residues on antifreeze and antilectin activities.
Structural requirements for antifreeze and antilectin activities of the antifreeze glycoproteins from the Antarctic fish Trematomus borckgrevinki have been compared. Graded removal of different amounts of the disaccharide side chain by p elimination caused similar reductions in the capacities for depressing the freezing temperature of water (antifreeze activity) and for inhibiting hemagglutinat...

متن کاملConvergent evolution of antifreeze glycoproteins in Antarctic notothenioid fish and Arctic cod.
Antarctic notothenioid fishes and several northern cods are phylogenetically distant (in different orders and superorders), yet produce near-identical antifreeze glycoproteins (AFGPs) to survive in their respective freezing environments. AFGPs in both fishes are made as a family of discretely sized polymers composed of a simple glycotripeptide monomeric repeat. Characterizations of the AFGP gen...
متن کاملNonhepatic origin of notothenioid antifreeze reveals pancreatic synthesis as common mechanism in polar fish freezing avoidance.
Phylogenetically diverse polar and subpolar marine teleost fishes have evolved antifreeze proteins (AFPs) or antifreeze glycoproteins (AFGPs) to avoid inoculative freezing by internalized ice. For over three decades since the first fish antifreeze (AF) protein was discovered, many studies of teleost freezing avoidance showed hepatic AF synthesis and distribution within the circulation as pivota...
