چکیده

Free-living Bradyrhizobiumjaponicum grown heterotrophically with 1 ,uM 63Ni2' accumulated label. Strain SR470, a Hupc mutant, accumulated almost 10-fold more 63Ni2+ on a per-cell basis than did strain SR, the wild type. Nongrowing cells were also able to accumulate nickel over a 2-h period, with the Hupc mutant strain SR470 again accumulating significantly more 63Ni2+ than strain SR. These results suggest that this mutant is constitutive for nickel uptake as well as for hydrogenase expression. The apparent Kms for nickel uptake in strain SR and strain SR470 were found to be similar, approximately 26 and 50 ,uM, respectively. The. V values, however, were significantly different, 0.29 nmol of Ni/min per 108 cells for SR and 1.40 nmol of Ni/min per 108 cells for SR470. The uptake process was relatively specific for nickel; only Cu2' and Zn2+ (10 ,uM) were found to appreciably inhibit the uptake of 1 ,uM Ni, while a 10-fold excess of Mg2+, Co2+, Fe3+, or Mn2+ did not affect Ni2+ uptake. The lack of inhibition by Mg2+ indicates that nickel is not transported by a magnesium uptake system. Nickel uptake was also inhibited by cold (53% inhibition at 4°C) and slightly by the ionophores nigericin and carbonyl cyanide m-chlorophenylhydrazone. Other ionophores did not appreciably affect nickel uptake, even though they signfficantly stimulated 02 uptake. The cytochrome c oxidase inhibitors azide, cyanide, and hydroxylamine did not inhibit Ni2+ uptake, even at concentrations (of cyanide and hydroxylamine) that inhibited 02 uptake. The addition of oxidizable substrates such as succinate or gluconate did not increase nickel uptake, even though they increased respiratory activity. Nickel uptake showed a pH dependence with an optimum at 6.0. Most (approximately 85%) of the 63Ni2+ taken up in 1 min by strain SR470 was not exchangeable with cold nickel.

Nickel resistance determinants in bradyrhizobium strains from nodules of the endemic New Caledonia legume Serianthes calycina.

Bradyrhizobium strains, isolated in New Caledonia from nodules of the endemic legume Serianthes calycina growing in nickel-rich soils, were able to grow in the presence of 15 mM NiCl2. The genomes of these strains harbored two Ni resistance determinants, the cnr and nre operons. By constructing a cnrA mutant, we demonstrated that the cnr operon determines the high nickel resistance in Bradyrhiz...

Dual roles of Bradyrhizobium japonicum nickelin protein in nickel storage and GTP-dependent Ni mobilization.

The hydrogenase accessory protein HypB, or nickelin, has two functions in the N(2)-fixing, H(2)-oxidizing bacterium Bradyrhizobium japonicum. One function of HypB involves the mobilization of nickel into hydrogenase. HypB also carries out a nickel storage/sequestering function in B. japonicum, binding nine nickel ions per monomer. Here we report that the two roles (nickel mobilization and stora...

Regulation of urease in Bradyrhizobium colonizing green gram (Vigna radiata (L.) Wilczek).

In the present study attempts have been made to characterize urease expression in slow growing Bradyrhizobium strains TAL442 and MO5 which are endosymbionts of green gram (Vigna radiata (L.)Wilczek). It was found that urease activity in vegetative cells of both the strains was inducible unlike their fast growing counterparts. Mode of regulation in TAL442 was governed by presence of ammonia. Ure...

Ability of some crops for phytoremediation of nickel and zinc heavy metals from contaminated soils

The present study was aimed at comparing the ability of three crop plants for phytoremediation of zinc (Zn) and nickel (Zn) from soils. A factorial (3×2×3) experiment based on RCBD was used to compare the treatments, and was repeated three times. The first factor was crop type (wheat, clover and rapeseed), the second factor was heavy metal types (zinc (Zn) and nickel (Ni)) and the third factor ...

TonB-dependent receptors in nitrogen-fixing nodulating bacteria.

TonB-dependent receptors (TBDRs) allow Gram-negative bacteria to uptake scarce resources from competitive environments with very high affinity. Early reports on TBDRs focused on the uptake of siderophore-iron complexes but recent studies have showed that the spectrum of ligands includes sugars, vitamins, heme, and other non-ferrous cations. To investigate the possible roles of TBDRs in nitrogen...
