چکیده
We investigated the effect of hemodynamic shear forces on the expression of adhesive molecules, E-selectin, and intercellular adhesion molecule-l (ICAM-1) on human umbilical vein endothelial cells (HUVEC) exposed t o laminar (8 dynes/ cmz) or turbulent shear stress 18.6 dyneslcrn' average), or to a static condition. Laminar flow induced a significant timedependent increase in the surface expression of ICAM-1, as documented by flow cytometry studies. Endothelial cell surface expression of CAM-1 in supernatants of HUVEC exposed t o laminar flow was not modified, excluding the possibility that HUVEC exposed t o laminar flow synthetize factors that upregulate ICAM-1. The effect of laminar flow was specific for ICAM-1, while E-selectin expression was not modulated by the flow condition. Turbulent flow did not affect surface expression of either E-selectin or ICAM-1. To evaluate the functional significance of the laminar-flow-induced increase in ICAM-1 expression, we studied the dynamic interaction of total leukocyte suspension with HUVEC ex-

Fluid shear stress modulates surface expression of adhesion molecules by endothelial cells.
We investigated the effect of hemodynamic shear forces on the expression of adhesive molecules, E-selectin, and intercellular adhesion molecule-1 (ICAM-1) on human umbilical vein endothelial cells (HUVEC) exposed to laminar (8 dynes/cm2) or turbulent shear stress (8.6 dynes/cm2 average), or to a static condition. Laminar flow induced a significant time-dependent increase in the surface expressi...
متن کاملLaminar shear stress elicit distinct endothelial cell E-selectin expression pattern via TNFα and IL-1β activation.
The ability to discriminate cell adhesion molecule expression between healthy and inflamed endothelium is critical for therapeutic intervention in many diseases. This study explores the effect of laminar flow on TNFα-induced E-selectin surface expression levels in human umbilical vein endothelial cells (HUVECs) relative to IL-1β-induced expression via flow chamber assays. HUVECs grown in static...

Leukocyte transmigration can be affected by shear stress; however, the mechanisms by which shear stress modulates transmigration are unknown. We found that adhesion of eosinophils or an eosinophilic cell line to intereukin 4-stimulated endothelial cells led to a shear-dependent increase in endothelial cell intracellular calcium and increased phosphorylation of extracellular signal-regulated kin...
متن کاملMechanical force modulates global gene expression and - catenin signaling in colon cancer cells
Introduction The effects of mechanical force induced by fluid shear stress on the activity of several signal transduction pathways in endothelial cells is well documented (for a review, see Resnick and Gimbrone, Jr, 1995). Mechanical forces, such as those generated by fluid shear stress, are sensed at the cell surface (at adhesion points) by 1 and 3 integrins, and transduced through the cell ...
متن کاملSpatial regulation of inflammation by human aortic endothelial cells in a linear gradient of shear stress.
OBJECTIVE Atherosclerosis is a focal disease that develops at sites of low and oscillatory shear stress in arteries. This study aimed to understand how endothelial cells sense a gradient of fluid shear stress and transduce signals that regulate membrane expression of cell adhesion molecules and monocyte recruitment. METHODS Human aortic endothelial cells were stimulated with TNF-alpha and sim...
