Highly efficient inoculum propagation in perfusion culture using WAVE Bioreactor™ systems

چکیده
Introduction A perfusion-based process was developed to increase the split ratio during the scale-up of CHO-STM cell cultures. Fedbatch cultures were inoculated with cells propagated in either batch or perfusion cultures. All cultures were grown in disposable CellbagTM bioreactors using the WAVE Bioreactor system. Cell concentrations of 4.8 × 10 cells/mL were achieved in the perfusion culture, whereas the final cell concentration in the batch culture was 5.1 × 10 cells/ mL. The higher cell concentration of the perfusion culture allowed for a more than six-fold increase of the split ratio to about 1:30. The method described here, can reduce the number of required expansion steps and eliminate the need for one or two bioreactors in the seed train. Singleuse bioreactors at benchtop scale can be used for direct inoculation of production bioreactors. Alternatively, high biomass concentrations accumulated in perfusion culture can be used to seed production vessels at increased cell concentrations. Thus, the process time in these bioreactors, which often is the bottleneck in plant throughput, can be shortened.

متن کاملGene transfection of HEK cells on supermacroporous polyacrylamide monoliths: a comparison of transient and stable recombinant protein expression in perfusion culture.
Transient and continuous recombinant protein expression by HEK cells was evaluated in a perfused monolithic bioreactor. Highly porous synthetic cryogel scaffolds (10 ml bed volume) were characterised by scanning electron microscopy and tested as cell substrates. Efficient seeding was achieved (94% inoculum retained, with 91-95% viability). Metabolite monitoring indicated continuous cell growth,...
متن کاملHigh yield of human monoclonal antibody produced by stably transfected Drosophila schneider 2 cells in perfusion culture using wave bioreactor.
Since it was first introduced in late 1990s Wave bioreactor has been used for protein production by mammalian and insect cell lines. However, using Wave bioreactor to produce human monoclonal antibody by stable Drosophila Schneider 2 (S2) cell transfectants has not been reported before. In this study, S2 cells were co-transfected with an inducible vector expressing human monoclonal antibody hea...
متن کاملBioreactor culture of oil palm (Elaeis guineensis) and effects of nitrogen source, inoculum size, and conditioned medium on biomass production.
We report the successful culture of oil palm (Elaeis guineensis Jacq.) suspension cells in a bioreactor. In vitro propagation of this perennial monocotyledonous tree is an important part of the oil palm industry's approach to clonal propagation of high-yielding accessions. During culture of oil palm cells in a batch bioreactor, nutrients and extracellular metabolites were monitored, and kinetic...
متن کاملOsteogenic Differentiation and Mineralization on Compact Multilayer nHA-PCL Electrospun Scaffolds in a Perfusion Bioreactor
Background: Monolayer electrospun scaffolds have already been used in bone tissue engineering due to their high surface-to-volume ratio, interconnectivity, similarity to natural bone extracellular matrix (ECM), and simple production. Objectives: The aim of this study was to evaluate the dynamic culture effect on osteogenic differentiation and mineralizationi into a compact cellular multilayer ...
