Lysozyme Lysis of Gram-negative Bacteria without Production of Spheroplasts.

چکیده

Cells of Gram-negative bacteria undergo lysis when treated with lysozyme in the presence of ethylenediaminetetraacetic acid (EDTA) and tris buffer, as shown by Repaske. However, contrary to the prevalent assumption, lysis is not necessarily preceded by formation of a spheroplast as the cell wall is damaged. Treatment of Escherichia coli and Pseudomolzas mruginosa with the lytic system was shown to cause the formation of osmotically fragile rods, rather than spheres. The extent of destruction of the cell walls of Gram-negative bacteria by lysozyme in this system is, at least in some cases, less than has been generally supposed.

Lysis of frozen and thawed cells of Escherichia coli by lysozyme and their conversion into spheroplasts.

Since the demonstration by Fleming (1922) of the lytic effects of lysozyme on Bacillus megaterium, this enzyme has been found to lyse a variety of bacteria. Most of the gram-positive bacteria, particularly Micrococcus lysodeikticus, Sarcina lutea, and B. megaterium, are very sensitive to the action of this enzyme; lysozyme completely removes their cell wall, leaving behind naked protoplasts whi...

Inactivation of gram-negative bacteria by lysozyme, denatured lysozyme, and lysozyme-derived peptides under high hydrostatic pressure.

We have studied the inactivation of six gram-negative bacteria (Escherichia coli, Pseudomonas fluorescens, Salmonella enterica serovar Typhimurium, Salmonella enteritidis, Shigella sonnei, and Shigella flexneri) by high hydrostatic pressure treatment in the presence of hen egg-white lysozyme, partially or completely denatured lysozyme, or a synthetic cationic peptide derived from either hen egg...

Localization of alkaline phosphatase in three gram-negative rumen bacteria.

Of the three species (Bacteroides ruminicola, B. succinogenes, and Megasphaera elsdenii) of anaerobic gram-negative rumen bacteria studied, only B. ruminicola produced significant amounts of alkaline phosphatase. This enzyme, which is constitutive, showed a greater affinity for p-nitrophenylphosphate than for sodium-beta-glycerophosphate and was shown to be located exclusively in the periplasmi...

Effect of polymyxin on the lysis of Neisseria catarrhalis by lysozyme.

In a previous communication (Warren et al., 1955) it was noted that gram-negative bacteria pretreated with heat or acetone were lysed by lysozyme. Since the bacteria studied were not sensitive to the enzyme when conventional methods were used, the possibility was considered that this pretreatment made the substrate in the cell more accessible to the action of the lysozyme by the removal or dest...

Lysis of Desulfovibrio vulgaris by ethylenediaminetetraacetic acid lysozyme.

The lysis of certain gram-negative bacteria by ethylenediaminetetraacetic acid (EDTA) and lysozyme was first reported by Repaske (7, 8). Lysis by EDTA alone was reported for Pseudomonas aeruginosa (3, 8), Vibrio succinogenes (9), and Alkaligenes faecalis (3). The mechanism for lysis by EDTA (2) in conjunction with lysozyme has been attributed to the interruption of the integrity of the cell sur...
