Storage and secretion processes in the spore of Gigaspora margarita Becker & Hall as revealed by high-pressure freezing and freeze substitution

چکیده
Spores of Gigaspora margarita Becker & Hall, an arbuscular mycorrbizal fungus, were cryofixed using bigbprcssure freezing and prepared for transmission electron microscopy by freeze substitution. I'hc cryotechniques improved tbe ultrastructural preservation of tbe spore cytoplasm and revealed storage and secretion processes in tbe germinating spores of G. margarita. The storage structures m the bydrated and/or germinating spores bave been identified as bflongtng to rhree differenr (-lasses: (i) lipid dropJeTs limited by a membrane, (ii) protein-like bodies inside specialized vacuoles, and (ii) ghcogen particles. During germ-tube differentiation, an abundant membranous system was obser\-ed. it consists of rougb KR. tubules and balloon-iike Golgi equivalents, wbicb produce vesicles associated witb electron-dense granules. Tbe granules seem (u be incorporated info the waU. The results suggest that secretion and storage occur in tbe fungal spore during events comparable to those in yeasts or plant cells.

Sperm freeze-drying; effects on sperm parameters
Today, sperm cryopreservation is a routine practice in infertility clinics. One of the alternative methods to preserve sperm is freeze-drying. This process consists of three steps. The first step is sample freezing, during which the solvent is separated from the solvent. Water forms ice crystals and the solute is placed between the ice crystals. Freezing may be done in a separate device or in a...
متن کاملPreservation of plasma membrane ultrastructure in Drosophila embryos and larvae prepared by high - pressure freezing and freeze - substitution
The ultrastructural analysis of biological structures is often critical to the understanding of biological processes. Fixation by freeze-substitution after immobilisation by High-pressure freezing (HPF-FS) is a powerful method to preserve the ultrastructure of small specimens such as the embryo of the fruit fly Drosophila melanogaster. But only a small fraction of specimens prepared following t...
متن کاملAn assessment of high-pressure freezing and freeze substitution protocols for cultured cells
Since the advent of cryo-electron microscopy (cryo-EM), the close-to-life and fine structure preservation of biological samples has become a realized procurement, surpassing the fixation quality and avoiding the numerous artifacts inherent to conventional chemical preservation methods. Cryo-EM techniques such as high-pressure freezing followed by freezesubstitution (HPF/FS) have facilitated the...
متن کاملUltrastructure of Fat Body Cells of Freeze-susceptible and Freeze-tolerant Eurosta solidaginis Larvae After Chemical Fixation and High Pressure Freezing
In response to low environmental temperatures third instar larvae of the goldenrod gall fly, Eurosta solidaginis, undergo the process of cold-hardening in which they acquire freeze-tolerance. Cold-hardening includes the elevation of the supercooling point, the temperature at which body water spontaneously freezes, and the accumulation of the low-molecular-mass cryoprotectants, glycerol, sorbito...
متن کاملNovel features of Brassica napus embryogenic microspores revealed by high pressure freezing and freeze substitution: evidence for massive autophagy and excretion-based cytoplasmic cleaning.
Induction of embryogenesis from isolated microspore cultures is a complex experimental system where microspores undergo dramatic changes in developmental fate. After ~40 years of application of electron microscopy to the study of the ultrastructural changes undergone by the induced microspore, there is still room for new discoveries. In this work, high pressure freezing and freeze substitution ...
