Wet splicing systems

چکیده
Splicing systems were originally developed as a mathematical or dry model of the generative capacity of DNA molecules in the presence of appropriate enzymes. The accuracy with which the model predicts the behavior of the corresponding biological or wet splicing system is investigated. A simple example of a wet splicing system is shown to generate in vitro the splicing language predicted by the corresponding dry splicing system.

Splicing to the Limit
We consider the result of a wet splicing procedure after the reaction has run to its completion, or limit, and we try to describe the molecules that will be present at this final stage. In language theoretic terms the splicing procedure is modeled as an H system, and the molecules that we want to consider correspond to a subset of the splicing language which we call the limit language. We give ...
متن کاملRole of Aberrant Alternative Splicing in Cancer
Alternative splicing can alter genome sequence and as a consequence, many genes change to oncogenes. This event can also affect protein function and diversity. The growing number of study elucidate the pathological influence of impaired alternative splicing events on numerous disease including cancer. Here, we would like to highlight the significant role of alternative splicing in cancer biolog...
متن کاملSodium Butyrate and Valproic Acid as Splicing Restoring Agents in Erythroid Cells of b-Thalassemic Patients
Background: b-Thalassemia is a common autosomal recessive disorder in human caused by a defect in b-globin chain synthesis. The most common mutations causing b-Thalassemia have been found to be splicing mutations. Most of which activate aberrant cryptic splicing/sites without complete disruption of normal splicing. IVSI-110 mutation, a common splicing mutation, leads to a 90% reduction of norma...
متن کاملAlternative RNA splicing: contribution to pain and potential therapeutic strategy
Since the sequencing of metazoan genomes began, it has become clear that the number of expressed proteins far exceeds the number of genes. It is now estimated that more than 98% of human genes give rise to multiple proteins through alternative pre-mRNA splicing. In this review, we highlight the known alternative splice variants of many channels, receptors, and growth factors that are important ...
متن کاملRestoration of correct splicing in IVSI-110 mutation of β-globin gene with antisense oligonucleotides: implications and applications in functional assay development
Objective(s): The use of antisense oligonucleotides (AOs) to restore normal splicing by blocking the recognition of aberrant splice sites by the spliceosome represents an innovative means of potentially controlling certain inherited disorders affected by aberrant splicing. Selection of the appropriate target site is essential in the success of an AO therapy. In this study, in search for a splic...
