چکیده

The formation of a soluble H(3)-thymidine derivative pool has been examined in Tetrahymena pyriformis as a function of macronuclear DNA synthesis during the cell life cycle. An autoradiographic technique which allows the detection of water-soluble materials within a cell has shown that these cells do not take up and retain exogenous H(3)-thymidine during G1 or G2. Uptake of H(3)-thymidine is restricted to the S period of the cell cycle. Additional autoradiographic experiments show, however, that a soluble pool of H(3)-thymidine derivatives persists from the end of one DNA synthesis period to the beginning of the next synthesis period in the subsequent cell cycle. Since this persisting pool cannot be labeled with H(3)-thymidine, the pool does not turn over during non-S periods.

The Distribution of Dna among Dividing Mitochondria of Tetrahymena Pyriformis

A squash technique was developed for log phase Tetrahymena pyriformis which permitted the resolution of over 100 individual mitochondria from a single cell. Mitochondria incorporated thymidine at all stages of the cell cycle, even when nuclear DNA synthesis was not occurring. During the stage of macronuclear DNA synthesis, however, there was a significant increase in the extent of mitochondrial...

Cell Division and Dna Synthesis in Tetrahymena Pyriformis Deprived of Essential Amino Acids

The question of amino acid requirements for DNA synthesis and cell division has been studied in Tetrahymena pyriformis by depriving cells of histidine and tryptophan at defined stages in the interdivision interval. Deprivation any time before DNA synthesis does not prevent the initiation of such synthesis but completely inhibits the following division and limits the increase in DNA, as measured...

Synthesis of Deoxyribonucleic Acid by Micro- and Macronuclei of Tetrahymena Pyriformis

Evidence as to the times of DNA synthesis in micronucleate Tetrahymena pyriformis (mating type II, variety 1) has been obtained by briefly exposing individuals of different ages to tritiated thymidine, returning them to non-radioactive medium, fixing at division, and preparing autoradiographs. A variable length of interphase, ranging from a few minutes to about 2 hours, has been found to preced...

Regulation of ribosomal RNA synthesis in Tetrahymena pyriformis.

Ribosomal RNA is synthesized at constant rate during most of the cell cycle in heat-shock synchronized populations of Tetrahymena pyriformis. Early in each macronuclear S-period the rate of synthesis increases abruptly, concomitant with replication of the genes coding for ribosomal RNA. The increase is prevented by inhibitors of DNA replication, added prior to the S-period. Similarly, in cultur...

Electron-microscopic observations on the macronuclear development of Stylonychia mytilus and Tetrahymena pyriformis (Ciliophora-Protozoa).

This report describes an ultrastructural investigation of macronuclear development following conjugation in Stylonycltia mytilus (a spirotrichous ciliate) and Tetrahymena pyriformis (a holotrichous ciliate). In S. mytilus, polytene chromosomes are formed in the young macronucleus (macronuclear Anlage). They are subsequently broken between the bands by 'membranous' partitions; the assembly of th...
