چکیده
naling pathway. For this, we focused on Pea3/Ets (Pea3, Erm, Er81), a subfamily of Ets transcription factors, since it has been suggested that Pea3/Ets is a direct transcriptional target of Fgf signaling. We first checked that Pea3 and Erm are expressed in the midbrain and the hindbrain around stage10 of the chick embryos. We then obtained results that Pea3 and Erm expressions are regulated by Fgf-Ras/ERK signaling; Pea3/Erm were induced after misexpression of Fgf8 and repressed by misexpression of dominant-negative form of Ras. In order to examine function of Pea3/Erm in the isthmus, we constructed dominant-negative form of Pea3 (Pea3-DN) and misexpressed it in the mesencephalon and the metencephalon at st.9 chick embryos. Surprisingly, Shh was ectopically induced at Pea3DN expressing sites in the midbrain and hindbrain. Oculomotor neurons that differentiate in the ventral midbrain differentiated ectopically in the dorsal side of the midbrain. It is speculated that Fgf signaling is involved in not only caudorostral axis formation, but also ventro-dorsal axis formation of the midbrain by repressing Shh through Pea3/Ets transcription factors.

Dynamic Sox5 protein expression during cranial ganglia development.
Sox5 is a member of the SoxD group of HMG-box transcription factors that, during the early stages of development, promotes neural crest generation. However, little is known about Sox5 function in neural crest derivatives such as the peripheral sensory nervous system. We have analysed the embryonic expression of Sox5 during chick cranial ganglia development, from the stages of ganglia condensati...
متن کاملProx1 physically and functionally interacts with COUP-TFII to specify lymphatic endothelial cell fate.
Specification of endothelial cell (EC) fate during vascular development is controlled by distinct key regulators. While Notch plays an essential role in induction of arterial phenotypes, COUP-TFII is required to maintain the venous EC identity. Homeodomain transcription factor Prox1 functions to reprogram venous ECs to lymphatic endothelial cells (LECs). Here, we report that the venous EC fate ...
متن کاملDistinct Requirements for Cranial Ectoderm and Mesenchyme-Derived Wnts in Specification and Differentiation of Osteoblast and Dermal Progenitors
The cranial bones and dermis differentiate from mesenchyme beneath the surface ectoderm. Fate selection in cranial mesenchyme requires the canonical Wnt effector molecule β-catenin, but the relative contribution of Wnt ligand sources in this process remains unknown. Here we show Wnt ligands are expressed in cranial surface ectoderm and underlying supraorbital mesenchyme during dermal and osteob...
متن کاملModifiers of muscle and heart cell fate specification identified by gain-of-function screen in Drosophila
The homeobox genes ladybird in Drosophila and their vertebrate counterparts Lbx1 genes display restricted expression patterns in a subset of muscle precursors and are both implicated in diversification of muscle cell fates. In order to gain new insights into mechanisms controlling conserved aspects of cell fate specification, we have performed a gain-of-function (GOF) screen for modifiers of th...
متن کاملSpecification of functional cranial placode derivatives from human pluripotent stem cells.
Cranial placodes are embryonic structures essential for sensory and endocrine organ development. Human placode development has remained largely inaccessible despite the serious medical conditions caused by the dysfunction of placode-derived tissues. Here, we demonstrate the efficient derivation of cranial placodes from human pluripotent stem cells. Timed removal of the BMP inhibitor Noggin, a c...
