چکیده

In an earlier investigation from this laboratory (I), prostatic phosphomonoesterase was used for the partition of the phosphomonoester and phosphodiester groups in ribonuclease digests. It was shown that the inorganic phosphate formed during incubation of ribonuclease digests with phosphatase originated practically exclusively from the acid-stable (pyrimidine) nucleotide groups of ribonucleic acids. It was concluded that the action of ribonuclease involved specifically pyrimidine nucleotide groups of yeast ribonucleic acids. Independent observations from several other laboratories likewise suggest the assumption that the action of ribonuclease involves specifically, or at least preferentially, the pyrimidine nucleotide groups of yeast ribonucleic acid (2-4). It is still unknown, however, how many of the total pyrimidine nucleotide groups of yeast ribonucleic acid are hydrolyzable by ribonuclease. This question is the problem of the present investigation.

A Practical Preparation of Ribonuclease-S and the Action of Subtilisin on Ribonuclease-B ‡

All preparations of ribonuclease-S so far reported have employed limited digestion of ribonuclease-A with subtilisin,' a bacterial proteinase from B. subtilis described by Giintelberg and Ottesen.' The original supply of this latter enzyme is now exhausted. Another sample of a crystalline proteinase from B. subtilis (NOVO Enzyme) has been shown to differ from the original in several respects,' ...

Ribonuclease A digestion by proteinase K.

The digestion of ribonuclease A by proteinase K yielded one major degradation product only, which could not be distinguished from ribonuclease S by electrophoretical and immunological methods. This component (ribonuclease K) possessing full catalytic activity was characterized to be (1--20/21--124) ribonuclease A. Combined action of proteinase K and trypsin on ribonuclease A leads to a signific...

The molecular mechanism of action of methylthioinosinedicarboxaldehyde on ribonuclease A as a model system.

The molecular mechanism of action of the antitumor drug, methylthioinosine periodate oxidation product, was demon strated. This periodate oxidation product (methylthioinosinedicarboxaldehyde) inhibited bovine pancreatic ribonuclease A. The inhibition was due to Schiffs base formation with e-amino groups of lysine residues, one of which was in close proximity to the catalytic center of the enzyme.

The interaction between ribonuclease and mononucleotides as measured spectrophotometrically.

The action of pancreatic ribonuclease is inhibited by a number of mononucleotides (l), of which one of the most effective is cytidine 2’-phosphate (2). We have presented evidence (3) that cytidine 2’-phosphate is a competitive inhibitor of the action of ribonuclease upon either ribonucleic acid or cytidine 2’) 3’ cyclic phosphate, but that the dissociation constant of the cytidine 2’-phosphate-...
