چکیده

The effects of some factors on the preparation and regeneration of protoplasts from the taxol-producing fungus Nodulisporium sylviforme were discussed in this paper, including combination of various enzymes, digesting time and temperature, and pH value, osmotic stabilizers, pretreation, culture medium and culture method. Under the condition of the mixed enzymes in osmotic pressure stabilizer (0.7 mol/L NaCl) combination of 3% lywallzyme, 2% snailase and 1% lysozyme, pH 5.5~6.0, digesting time 9h and digesting temperature 30°C, the results showed that the highest preparation frequency of protoplasts is a concentration of 2.0×10 protoplasts per microlitre. After the obtained protoplasts were purified, they would be regenerated by bilayer plate culture method in PDA regenerative medium. This study laid the foundation to construct the engineering taxol-producing strains by protoplast mutagensis, transformation, fusion, and turn to a new page of research of producing anti-tumor drug taxol by microbiological fermentation. Regeneration of yeast protoplasts prepared using Novozym 234.

Novozym 234 (Novo lndustri A/S, Bagsraerd, Denmark) is a comparatively cheap and highly effective commercial preparation, derived from the fungus 7richoderma harziunum, which is used for the enzymic degradation of the cell wall in many yeasts and filamentous fungi in order to release protoplasts [ 1 , 21. The factors involved in protoplast isolation using this system have been investigated by C...

A Review on Taxol Production through Biotechnological Approaches

Taxol is a very important anticancer drug which was first isolated from Yew plant (Taxus spp.). However, Taxol supply by extraction from natural sources, has several limitations, and cannot meet current market's demands. Therefore, it seems necessary to use alternative production methods. Producing Taxol through biotechnological approaches is among the main options which have some advantages su...

PLANT REGENERATION FROM MESOPHYLL PROTOPLASTS OF POTATO (SOLANUM TUBEROSUM L.) CULTIVAR DELAWARE USING SILVER THIOSULFATE (STS)

The use of in vitro potato shoot cultures for protoplast isolation is desirable for consistent protoplast qualities. Ethylene build-up in the culture vessels causes problems for leaf growth (e.g. decreasing leaf surface, dry weight and chlorophyll) in shoot culture. Fifty to one hundred µM Silver thiosulfate (STS) produced a larger leaf area as a source of plant material for protoplast isolatio...

SHOOT REGENERATION FROM SAFFRON PROTOPLASTS IMMOBILIZED IN Ca-ALGINATE BEADS

Saffron (Crocus sativus L.) protoplasts were isolated from the cells of a suspension culture or calli with a solution of Cellulase, Pectinase and Hemicellulase and embedded in Ca-alginate beads. They were cultured with or without nurse cells in MS medium supplemented with 2,4-D and 6-benzylaminopurine at 25°C. After several changes of medium, cell-clusters appeared on the surface of the Ca-algi...
