چکیده

The effect of fluridone on the growth of plants and drought stressinduced abscisic acid (ABA) accumulation were studied in mature leaf tissue of Vicia faba L. plants grown under different light intensity. Drought stress was induced by allowing the leaves to lose 12% of fresh weight. Treatment of Vicia plants (grown at high light intensity – 600μmol.m-2.s-1) with 10μmoles fluridone caused a considerable decrease in the rate of growth and bleaching of the plants. Fluridone fully blocked stress-induced ABA accumulation. Plants grown under low light intensity (40μmol.m-2.s-1) had diminished level of ABA after impossition the leaves to dehydration. Fluridone treatment reduced the level of ABA in unstressed as well as dehydrated leaves. The accumulation of ABA in 21-day-old green Vicia plants was considerably reduced when they were exposed to dark periods (24h, 48h, and 72h) just before the impossition of stress. Twenty four hours after dark treatment dehydration of these leaves resulted in a 3 fold decrease in stress-induced ABA accumulation. Seventy two hours after dark treatment the level of stress-induced ABA was approximately equal to the prestressed values. Fluridone-treated plants failed to accumulate ABA when water stressed. The presented results are discussed in term with the role of light and fluridone on stress-induced ABA accumulation.

Water-stress-induced changes in the abscisic acid content of guard cells and other cells of Vicia faba L. leaves as determined by enzyme-amplified immunoassay.

A highly sensitive, solid-phase, enzyme-amplified immunoassay for the plant growth regulator (+)-abscisic acid (ABA) was developed. The assay sensitivity (0.2-10 fmol) was sufficient for analyzing free ABA in homogeneous tissue samples dissected from Vicia faba L. leaves. Eight hours after detached leaves had been desiccated to 10% loss of fresh weight, the bulk leaf ABA content increased from ...

The nitric oxide production and NADPH-diaphorase activity in root tips of Vicia faba L. under copper toxicity

Copper (Cu) toxicity on plants has become a major problem with increasing agricultural and environmental pollution. The effect of Cu on NADPH-diaphorase (NADPH-d) activity, commonly employed as a marker for nitric oxide synthase (NOS) activity and nitric oxide (NO) production, was investigated in root tips of Vicia faba L. We found that Cu stress stunted root growth and development of root hair...

Modulation Role of Abscisic Acid (ABA) on Growth, Water Relations and Glycinebetaine Metabolism in Two Maize (Zea mays L.) Cultivars under Drought Stress

The role of plant hormone abscisic acid (ABA) in plants under drought stress (DS) is crucial in modulating physiological responses that eventually lead to adaptation to an unfavorable environment; however, the role of this hormone in modulation of glycinebetaine (GB) metabolism in maize particularly at the seedling stage is still poorly understood. Some hydroponic experiments were conducted to ...

Bacillus licheniformis SA03 Confers Increased Saline–Alkaline Tolerance in Chrysanthemum Plants by Induction of Abscisic Acid Accumulation

Soil saline-alkalization is a major abiotic stress that leads to low iron (Fe) availability and high toxicity of sodium ions (Na+) for plants. It has recently been shown that plant growth promoting rhizobacteria (PGPR) can enhance the ability of plants to tolerate multiple abiotic stresses such as drought, salinity, and nutrient deficiency. However, the possible involvement of PGPR in improving...

Improved cold tolerance in Elymus nutans by exogenous application of melatonin may involve ABA-dependent and ABA-independent pathways

Melatonin is an important secondary messenger that plays a central role in plant growth, as well as abiotic and biotic stress tolerance. However, the underlying physiological and molecular mechanisms of melatonin-mediated cold tolerance, especially interactions between melatonin and other key molecules in the plant stress response, remain unknown. Here, the interrelation between melatonin and a...
