Modification of acetylcholine sensitivity of neuronal membrane by presynaptic stimulation.

چکیده
The action of electrical stimulation of one of the pallial nerves on the sensitivity of the bursting RPa1 neuron of Helix pomatia to acetylcholine (ACh) was investigated. The depolarizing effect of ACh was significantly decreased by presynaptic stimulation. Stimulation leads also to an attenuation of the ACh-induced increase in membrane conductivity. The effect of stimulation on the ACh evoked response of the membrane was reversibly blocked by cold and was completely eliminated after long term incubation of the neuron under "in vitro" conditions.

Botulinum toxin type A blocks the morphological changes induced by chemical stimulation on the presynaptic membrane of Torpedo synaptosomes.
The action of botulinum neurotoxin on acetylcholine release, and on the structural changes at the presynaptic membrane associated with the transmitter release, was studied by using a subcellular fraction of cholinergic nerve terminals (synaptosomes) isolated from the Torpedo electric organ. Acetylcholine and ATP release were continuously monitored by chemiluminescent methods. To catch the membr...
متن کاملENDOGENOUS RELEASE OF OPIATES BY REPETITIVE ELECTRICAL FIELD STIMULATION IN THE GUINEA-PIG AND RAT ILEAL LONGITUDINAL MUSCLE
The effect of repetitive electrical field stimulation and the response of the guinea-pig and rat ileal longitudinal muscle to single pulse stimulations was examined. Single pulse field stimulation produced twitch contraction which was inhibited by repetitive field stimulation (10 Hz, 40V, 0.5 msec for 5 m). This inhibition was largely, though never completely, reversed by naloxone. Contrac...
متن کاملVesicular localization and activity-dependent trafficking of presynaptic choline transporters.
Presynaptic synthesis of acetylcholine (ACh) requires a steady supply of choline, acquired by a plasma membrane, hemicholinium-3-sensitive (HC-3) choline transporter (CHT). A significant fraction of synaptic choline is recovered from ACh hydrolyzed by acetylcholinesterase (AChE) after vesicular release. Although antecedent neuronal activity is known to dictate presynaptic CHT activity, the mech...
متن کاملPresynaptic Type III Neuregulin1-ErbB signaling targets α7 nicotinic acetylcholine receptors to axons
Type III Neuregulin1 (Nrg1) isoforms are membrane-tethered proteins capable of participating in bidirectional juxtacrine signaling. Neuronal nicotinic acetylcholine receptors (nAChRs), which can modulate the release of a rich array of neurotransmitters, are differentially targeted to presynaptic sites. We demonstrate that Type III Nrg1 back signaling regulates the surface expression of alpha7 n...
متن کاملSynaptic Vesicle Depletion and Recovery in Cat Sympathetic Ganglia Electrically Stimulated in Vivo
This study examined the ultrastructure of presynaptic terminals after short periods of vigorous acetylcholine (ACh) secretion in the cat superior cervical ganglion in vivo. Experimental trunks of cats anesthetized with chloralose-urethane were stimulated supra-maximally for periods of 15-30 min and at several frequencies including the upper physiological range (5-10 Hz). Stimulated and contrala...
