Characteristics of cellobiose phosphorylase.

چکیده
The phosphorolysis of cellobiose was first demonstrated by Sih and McBee (1955a, b). These workers found that cellobiose phosphorylase was present in cell-free extracts of Clostridium thermocellum. Recently cellobiose phosphorylase has been reported from two other bacteria, Ruminococcus flavefaciens (Ayers, 1958, 1959), and Cellvibrio gilvus (Hulcher and King, 1958). This enzyme also catalyzes the synthesis of cellobiose froin a-D-glucose 1-phosphate and glucose (Sih, Nelson, and McBee, 1957; Ayers, 1959). In the present work, the stoichiometry of the reaction and the effect of physical and chemical agents on the activity of the enzyme were studied. Studies on the specificity of the enzyme revealed that D-glucose, D-xylose, L-xylose, 2-deoxyglucose, D-glucosamine, arsenate, and inorganic phosphate can act as substrates. Evidence has been obtained which indicates that the reaction proceeds by a single displacement mechanism. A preliminary report of this work has been published (Alexander, 1958).

Overcoming inefficient cellobiose fermentation by cellobiose phosphorylase in the presence of xylose
BACKGROUND Cellobiose and xylose co-fermentation holds promise for efficiently producing biofuels from plant biomass. Cellobiose phosphorylase (CBP), an intracellular enzyme generally found in anaerobic bacteria, cleaves cellobiose to glucose and glucose-1-phosphate, providing energetic advantages under the anaerobic conditions required for large-scale biofuel production. However, the efficienc...
متن کاملCloning and characterization of the glucooligosaccharide catabolic pathway beta-glucan glucohydrolase and cellobiose phosphorylase in the marine hyperthermophile Thermotoga neapolitana.
Characterization in Thermotoga neapolitana of a catabolic gene cluster encoding two glycosyl hydrolases, 1,4-beta-D-glucan glucohydrolase (GghA) and cellobiose phosphorylase (CbpA), and the apparent absence of a cellobiohydrolase (Cbh) suggest a nonconventional pathway for glucan utilization in Thermotogales. GghA purified from T. neapolitana is a 52.5-kDa family 1 glycosyl hydrolase with optim...
متن کاملSynthesis of [C]Cellobiose with Clostridium thermocellum Cellobiose Phosphorylase.
Cellobiose labeled either at the reducing end (4-O-beta-d-glucopyranosyl-d-[U-C]glucopyranose) or at the nonreducing end (4-O-beta-d-[U-C]glucopyranosyl-d -glucopyranose) was synthesized with Clostridium thermocellum cellobiose phosphorylase at greater than 98% purity. The radioactive cellobioses were identical to authentic cellobiose in melting point, optical rotation, isotopic dilution, and c...
متن کاملStructural dissection of the reaction mechanism of cellobiose phosphorylase.
Cellobiose phosphorylase, a member of the glycoside hydrolase family 94, catalyses the reversible phosphorolysis of cellobiose into alpha-D-glucose 1-phosphate and D-glucose with inversion of the anomeric configuration. The substrate specificity and reaction mechanism of cellobiose phosphorylase from Cellvibrio gilvus have been investigated in detail. We have determined the crystal structure of...
متن کاملCellodextrin phosphorylase from Ruminiclostridium thermocellum: X-ray crystal structure and substrate specificity analysis
The GH94 glycoside hydrolase cellodextrin phosphorylase (CDP, EC 2.4.1.49) produces cellodextrin oligomers from short β-1→4-glucans and α-D-glucose 1-phosphate. Compared to cellobiose phosphorylase (CBP), which produces cellobiose from glucose and α-D-glucose 1-phosphate, CDP is biochemically less well characterised. Herein, we investigate the donor and acceptor substrate specificity of recombi...
