چکیده

K+ channels play a key role in cellular physiology by regulating the efflux of K+ ions. They are the most diverse group of ion channel proteins; more thm 30 K+ channel genes have been characterized. RegulaOed by ATP, voltage, and calcium, multiple K+ channels coexist in the P-cell to regulate membrane potential, cell excitability, and insulin secretion. Recent developments at the molecular level have greatly expanded our lpnderstanding of P-cell K+ channel structure and function, especially in regard to the ATP-sensitive K+ channdl, the target for sulfonylurea drugs. Mutations in K+ channel genes underlie diseases as diverse as persistent hyperinsulinemia of infancy, cardiac long QT syndrome, cerebellar degeneration, and certain atauias. These discoveries have identified new pharmacological targets for pobisible therapeutic intervention in the treatment of diabetes. Diabetes 45: 845-853,1996

Perspectives in Diabetes Diabetes and Insulin Secretion The ATP-Sensitive K Channel (KATP) Connection

The ATP-sensitive K channel (KATP channel) senses metabolic changes in the pancreatic -cell, thereby coupling metabolism to electrical activity and ultimately to insulin secretion. When KATP channels open, -cells hyperpolarize and insulin secretion is suppressed. The prediction that KATP channel “overactivity” should cause a diabetic state due to undersecretion of insulin has been dramatically ...

Perspectives in Diabetes Diverse Roles of KAT P Channels Learned From Kir6.2 Genetically Engineered Mice

The regulation of insulin secretion from pancreatic -cells depends critically on the activities of their plasma membrane ion channels. ATP-sensitive K c h a nnels (KAT P channels) are present in many cells and regulate a variety of cellular functions by coupling cell metabolism with membrane potential. The activity of the KAT P channels in pancreatic -cells is regulated by changes in the ATP an...

New Perspectives on the Role of Hyperglycemia, Free Fatty Acid and Oxidative Stress in B-Cell Apoptosis

Apoptosis is a complex network of biochemical and molecular pathway with fine regulatory mechanisms that control the death event during several pathological situations in multi cellular organisms. It is the part of normal development that occurs in a variety of diseases and is known as aberrant apoptosis. Pancreatic β cell apoptosis is also a pathological feature which is common in both type 1 ...

K(ATP) channels and pancreatic islet blood flow in anesthetized rats: increased blood flow induced by potassium channel openers.

K(ATP) channels are important for insulin secretion and depolarization of vascular smooth muscle. In view of the importance of drugs affecting K(ATP) channels in the treatment of diabetes, we investigated the effects of these channels on splanchnic blood perfusion in general and pancreatic islet blood flow in particular. We treated anesthetized Sprague-Dawley rats with the K(ATP) channel opener...

Transgenic mouse overexpressing syntaxin-1A as a diabetes model.

Soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein receptor (SNARE) protein syntaxin-1A (STX-1A) plays a role not only in exocytosis, but also binds and regulates Ca(2+) and K(+) (voltage-gated K(+) and ATP-sensitive K(+) channels) to influence the sequence of events leading to secretion. Islet levels of STX-1A and cognate SNARE proteins are reduced in type 2 diabetic rodents, s...
