چکیده

Epigenetics describes a phenomenon of modifications in gene expression caused by heritable, but potentially reversible, alterations in the chromatin structure, DNA methylation, and post-transcriptional effects of small noncoding (micro) RNAs (miRNAs), without changes in the DNA sequence. These mechanisms represent novel targets for polyphenols and other natural products in the prevention and treatment of cancer and other diseases. This chapter summarizes the influence of various classes of polyphenol on the enzymatic activities of major enzymes involved in epigenetic gene regulation: DNA and histone methyltransferases (DNMTs and HMTs), histone acetyltransferases (HATs), histone deacetylases (HDACs), and histone demethylases (HDMs). Inhibitory effects mediated by (−)-epigallocatechin 3-gallate (ECGC) from green tea and the isoflavone genistein from soy in vitro and in vivo are described in detail. Functional consequences on gene expression in cell culture, in rodent models and in human intervention studies, are highlighted.

Plant phenolics in the prevention and treatment of cancer.

Epidemiological studies indicate that populations consuming high levels of plant derived foods have low incidence rates of various cancers. Recent findings implicate a variety of phytochemicals, including phenolics, in these anticancer properties. Both monophenolic and polyphenolic compounds from a large variety of plant foods, spices and beverages have been shown to inhibit or attenuate the in...

The Role of Epigenetics in Cancer Drug Resistance

Cancer is caused by aberrant genetic and epigenetic changes in genes expression. DNA methylation, histone modification, and microRNAs gene deregulation are the most known epigenetic changes in different stages of cancer. Since every tumor has its own specific epigenome, any abnormal pattern is a potential biomarker for classification of different types of tumors. Despite, tumorigenesis, abnorma...

Epigenetics in diagnosis of colorectal cancer

Colorectal cancer (CRC) is a third most common epithelial carcinoma. CRC is known to develop from the early precancerous lesion to full blown malignancy via definite phases due to cumulative mutations and aberrant methylation of number of genes. The use of serum biomarkers that is non-invasive to discriminate cancer patients from healthy persons will prove to be an important tool to improve the...
